Abstract


Objective: endothelial cells (endothelial cell, EC) for the maintenance of the vascular wall plays an important role in a stable environment, the integrity of vascular thrombosis EC, leukocyte adhesion and invasion, and growth inhibition of cytokine production and other important barrier. Percutaneous coronary intervention (percutaneous transluminal intervention, PCI) after intimal injury resulting not only prone to platelet adhesion and aggregation, and can release a variety of vasoactive substances, promote vasoconstriction and vascular smooth muscle cells (vascular smooth muscle cell, VSMC) proliferation and migration. Timely repair of damage can be reduced intimal VSMC migration to the intima, delaying vascular restenosis (Restenosis, RS) formation. Prevention of restenosis after PCI in addition to effectively inhibit the proliferation of VSMC, the early resumption of EC activity and accelerate the repair of EC, is the key to prevention and treatment of postoperative RS. Corilagin (Corilagin) from Euphorbiaceae 1a herb Phyllanthus (Phyllanthus urinaria) polyphenolic extract isolated monomeric compounds. Studies have suggested corilagin atherosclerosis prevention role. This research concept Chakelila Beijing vitro porcine coronary artery endothelial cell proliferation and to explore its RS and arteriosclerosis in the fight against the possible mechanism. Methods: The combination of mechanical digestion of isolated porcine coronary artery endothelial cells (porcine coronary endothelial cell, PCEC), and the endothelial cell culture medium (Endothelial Cell Medium, ECM) in the culture. Factor Ⅷ related antigen using immunofluorescence and electron microscopy identified endothelial cells, 4-8 cells for drug experiments. Trypan blue exclusion assay corilagin effects on cell survival; cell count and MTT assay corilagin right PCEC cell proliferation, make plans and calculate the effective half corilagin inhibitory concentration (IC50); flow cytometry corilagin detected on cell cycle of PCEC. Results: The cultured cells were PCEC, factor Ⅷ related antigen in immunofluorescence and cell fusion under an inverted microscope monolayer growth, cobblestone-like arrangement, seen under TEM WP body. 50 ~ 1000μmol / L of corilagin incubated with PCEC 24-96 hours after corilagin make PCEC reduced survival, and there was a certain degree of concentration and time dependent, IC5o of 244.7μmol / L . Flow cytometry shows that PCEC medication Houkelila Beijing's Go / G, phase fraction increased significantly; S phase and G2 / M phase cell percentage is also reduced, as compared with the control group, there was significant statistical significance (P lt; 0.01), and with the increase of drug concentration, the role will be further strengthened. Conclusion: corilagin right PCEC no significant role in promoting cell proliferation, and its mechanism may be related to the inhibition of cell cycle PCEC G1/G0 phase to S phase transition, so that the cell cycle arrest at G1/G0 phase related to proved corilagin on endothelial regeneration and repair without protection.
