Malaria is caused by a parasite , is one of the most serious infectious diseases , causing 100 million deaths annually , currently 109 countries are suffering from malaria infection. Malaria is transmitted by the Anopheles with Plasmodium bites but will inject human blood circulation in the body , it can lead to hypoglycemia, fever, anemia and other symptoms . There are four people and four kinds of malaria mouse malaria, which is the study of the summer Plasmodium falciparum mouse model best . Current research and development for vaccines against malaria There are many types , subunit vaccines are the most studied . It can cause a strong humoral immunity, cellular immunity but is weak , can not cause long-lasting protective immunity . Subunit vaccines require adjuvants to enhance the immune response auxiliary . MSP1 is the most important candidate subunit vaccine can cause a protective force . It cuts through a series of C-terminal 19kDa last only connected via GPI insect surface and into erythrocytes . Have previously reported findings from the supernatant of cultured Plasmodium certain substances malaria -infected patients with positive serum taking effect. Therefore, this study, we wanted to study the culture supernatant common parasite MSP1 and immune effects. First, the use of E. coli expression system to obtain summer Plasmodium strains AS MSP1 protein C-terminal 11kDa; followed short-term culture in vitro metabolic secretions obtained Plasmodium summer (ES). Secretion and metabolism of the MSP1 joint immunization of mice , detecting infection , ELISA detection antibody titer ; splenocytes from immunized mice to study the secretion of cytokines and cell activation. It was found that : (a) MSP1 to ES as adjuvant can effectively inhibit blood worms level ; produce higher levels of antibodies , mainly IgG1, IgG2c and IgG2b; (b) activation of dendritic cells, CD40, CD86 molecule increased expression , MHC-II expression was decreased ; (c) activation of CD4 T cells to the surface molecule CD62L expression was decreased ; (d) -induced lymphocyte proliferation , resulting in a high IFN-γ, IL-12 and IL- 10 . This study is the first to metabolic secretions as novel adjuvants . Our findings reveal the complex mechanisms of anti-malarial immunity induced and may malaria vaccine development is of great significance .
