Abstract


With the improved living standards of our people and a change in eating habits, cardiovascular events, and cerebrovascular events has become a major cause of death in China, and coronary heart disease is a serious impact on human health, cardiovascular disease morbidity and mortality tops of various diseases in the forefront , has now become one of the diseases that endanger the safety of human life in the world. Atherosclerosis (ATHEROSCLEROSIS, AS) is a the basic pathological basis of coronary heart disease. Since the AS the pathogenesis remains unclear, thus establishing the significance of a lipid metabolism and plaque similar human experimental AS whole animal model. Currently, many scholars widely used in the high-fat diet AS animal models, also found in previous animal experiments, high fat diet for a longer period of rabbits AS model established, individual animals in the laboratory using relatively small quail manufacturing high fat model of hyperlipidemia and arterial atherosclerosis experimental study, the model has individual small laboratory animals, simple operation, short experimental period, likely to be successful modeling. Objective: extraction and separation of four the Coptis alkaloids study its impact on hyperlipidemia and atherosclerosis. : Coptis dried rhizomes ethanol extracted by the acid-soluble plus alkali adjust PH macroporous resin total alkaloids, another part of the extraction, over silica gel column was obtained by recrystallization of berberine coptisine palmatine, and through HPLC detection of its purity, it is found that the purity of 90% or more. Quail hyperlipidemia and atherosclerosis model with exogenous cholesterol and high fat diet, divided into 7 groups: normal control group, ② model group, (3) fat veins Kang group, berberine, coptisine, Alabama Ting total alkaloid group, in addition to the normal group was fed a normal diet, and the rest of the group fed the high fat diet (normal diet of 79%, 14% lard, 6% peanut oil 1% cholesterol. berberine coptisine palmatine total alkaloids and the positive drug Zhimaikang continuous gavage for 4 weeks, serum cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) content ; eighth week of the experiment weighed to calculate the coefficient of the liver, do the aorta and liver biopsy for histological examination of light microscopy. mechanism, inhibition of HMG-CoA reductase activity using HPLC detection vitro compounds. Results: model compared, berberine, coptisine, and palmatine significantly reduce TC, TG and LDL-C, the three TC were reduced 35.6%, 32.5%, 26.8% and TG were reduced by 51.5%, 53.1% , 47.6%, LDL-C was reduced by 39.2%, 42.3%, 26.7%; pathological results showed that the severity of aortic atherosclerotic plaque can significantly reduce the Coptis alkaloids inhibit HMG-COA, but the effect was not significant Conclusion: Coptis alkaloids significantly lowering blood pressure and inhibit atherosclerosis role, mechanisms need further study.
