Abstract


[Objectives]:To establish the fingerprint study method of lotus leaves and to develop a HPLC method for determination of rutin and nuciferine in lotus leaves. discussing influence of lotus leaves quality from different places and different collected time. And to study lotus leaf extract in viro antioxidant activity.[Methods]:Different origins and harvesting time on the lotus leaf, after ultrasonic extraction, components were separated and its content was determinated by HPLC-PDA method. Developed by the State Pharmacopoeia Commission of the similarity of chromatographic fingerprint evaluation system of version (2004A), every spectrometry and data were analysised and calculation of similarity. After investgating on different antioxidant models, antioxidant capacity in vitro of the extracts made by different solvents were calculated. The results were compared with that of vitamin C.[Results]:High-quality chromatographic fingerprint of lotus leaves was obtained and 11 common fingerprint peaks were detected and the structures of three ones were identified by comparison with authentic standard. Similarity comparison was conducted among the samples from different origin and also the samples with different harvest time. There existed the more differences between the contents of rutin and nuciferine as the leaves were from the different places and different collecting time, in which the peak areas of active components in lotus leaves from Yueyang, Jiangsu, Zhejiang and Xiangtan were rather higher.The antioxidant activity in vitro of ethanol extract was the highest and water extract followed.[Conclusions]:The proposed method was characterized with simple operation procedure and good repeatability, indicating this method can be effectively used for identification and quality control of lotus leaves. There exists a certain the differences of components of different origins and harvesting time of the lotus leaf due to soil or climate and other factors. The results can be used to screen fine producting areas and guide the reasonable collecting and making the most of lotus leaves resources. The extracts of lotus leaves has affirmatory antioxidant activities in vitro.The strengths of antioxidant activity of the extracts made by different solvents have different significance.
