Abstract

Objective: To observe the injection verapamil on intestinal ischemia - reperfusion injury in the role of the intestinal mucosa, and to explore its mechanism. METHODS: The experiment in 21 healthy male SD rats were randomly divided into 3 groups (n = 7): the sham group (A), ischemia-reperfusion group (I / R group) (B group) and verapamil The intervention group (C). Rat mesenteric artery occlusion the 45min and open 2h copy intestinal ischemia - reperfusion injury model verapamil group in reperfusion 10min before slowly injected via the tail vein 0.5g / L verapamil injection 1mg / kg, reperfusion 2h after the animals were killed, to take a group of small intestine HE staining small intestinal pathological damage Chiu's score, Parallel immunohistochemistry (SP enzyme linked immunosorbent assay) detection of TNF-α, Bax, Bcl-2, further applications Microscopic image analysis system (Image Pro Plus) to detect its expression. Results: 1. Sham group small intestinal villus epithelial cells arranged in neat rows, no significant inflammatory cell infiltration and telangiectasia, congestion, the pathological damage Chiu's score 0.57 ± 0.35, TNF-α, Bax and Bcl-2 expression optical density (OD ) values ??were 0.084 ± 0.008,0.112 ± 0.013 and 0.297 ± 0.012; small intestinal ischemia-reperfusion group organizational structure severely damaged, fluff the top part of the damaged lamina propria inflammatory cell infiltration, the pathological damage Chiu's score 3.29 ± 0.57, TNF -α, Bax and Bcl-2 expression optical density (OD) values ??were 0.219 ± 0.012,0.290 ± 0.013 and 0.106 ± 0.008; ischemia-reperfusion group of small intestinal tissue injury Chiu's score, TNF-α, Bax expression compared with the sham group (P lt; 0.01) significantly increased the expression of Bcl-2 significantly decreased (P lt; 0.01). 2. Verapamil group of small intestinal mildly dilated capillaries, congestion, partly subcutaneous gap increases accompanied on cortical lamina propria moderate separation, the pathological damage Chiu's score 2.05 ± 1.23, TNF-α, Bax and Bcl -2 expression optical density (OD) values ??were 0.121 ± 0.008,0.200 ± 0.010 and 0.205 ± 0.008, and ischemia-reperfusion group (I / R group) compared to alleviate the structural damage of the small intestinal mucosa, pathological damage Chiu Karnofsky score, TNF-α, Bax expression significantly decreased (P lt; 0.01), Bcl-2 expression was significantly increased (P lt; 0.01). Conclusion: 1. Ischemia and reperfusion can cause visible damage of the small intestinal mucosa. Calcium channel blockers verapamil can significantly reduce intestinal ischemia reperfusion due to the degree of pathological damage of the small intestinal mucosa. Calcium channel blockers verapamil on intestinal ischemia - reperfusion injury possible mechanisms: ① inhibited cell proinflammatory mediators TNF-α release, prevent reperfusion apoptosis and necrosis; ② prevent intracellular calcium overload, lowered promote apoptotic protein Bax, raised the anti-apoptotic protein Bcl-2 expression, thereby inhibiting apoptosis of small intestinal.
