The Effect of Lipopolysacchride from Periodontal Pathogen Porphyromonas Gingivalis on the Inflammatory Factors in Tissue-specific Monocytes/macrophages

Objective To study the potential mechanisms of periodontal disorders on systemetic diseases, this study was aimed to compare the effect of porphyromonas gingivalis lipopolysacchride on the inflammatory factors (C-reaction protein, interlenkin-1beta, interlenkin-6, interlenkin-8, tumor necrosis factor-alpha) in different part of monocy- tes/macrophages from the same individual. The effect of exogenous interlenkin-10 was also investigated.Methods New Zealand rabbits were fed the high-fat diet for six weeks to induce hyperlipemia by measuring the blood lipids, monitoring weight, aortic pathology and hepatic pathology. Monocytes/macrophages were isolated from different parts of the normal and hyperlipemia rabbits,including peripheral mononuclear cells, alveolar macrophages, peritoneal macrophages and kupffer cells. The monocytes/macrophages were stimulated with porphyromonas gingivalis lipopolysacchride, Escherichia coli Lipopolysacchride and interlenkin-10. The gene expression of inflammatory factors were analysed and compared through the quatitaive realtime PCR method.Results After feeding with six weeks of high-fat diet, the blood lipids levels were higher in the high-fat diet group than in the control group (P﹤0.01). There were no difference between the two groups on body weight (P﹥0.05). By means of optical microscope, there were no difference on tendarterium between these two groups, however, the liver was partial fatty degeneration in the high-fat diet group with foam cells detected. The results of inflammatory gene expression showed: (1) The different part of monocytes/macrophages showed different response of inflammatory factors to the stimulation of LPS from porphyromonas gingivalis(P﹤0.05), indicating a site specific react. (2) 1μg/ml Pg-LPS on different part of monocytes/macrophages induced higher mRNA expression of inflammatory factors (CRP、IL-1β、IL-6、IL-8、 TNF-α) in the high-fat diet group compared with the control group(P﹤0.05). (3) 0.1μg/ml IL-10 could reduce the gene expression of LPS-induced,inflammatory factors (CRP、IL-1β、IL-6、IL-8、TNF-α) in monocytes/macrophages(P﹤0.05).Conclusion This study has successfully established hyperlipemia animal model with New Zealand rabbits after six weeks high-fat diet. Site specific reaction of different part monocytes/macrophages stimulated to Pg-LPS was identified. The gene expression of inflammatory factors in high-fat diet group showed a stronger reaction compared with the control group. Periodontal pathogen Pg-LPS could increase the gene levels of inflammatory factors in different part monocytes/macrophages. IL-10 could inhibit the expression of inflammatory factors in different part monocytes/ macrophages.
