Corneal neovascularization (corneal neovascularization, CNV) can lead to structural damage eye tissue damage and visual function is one of the major blinding. Experiments confirmed that vascular endothelial growth factor (vascular endothelial growth factor, VEGF) in corneal neovascularization plays an important role in the process. In recent years, Avastin has been reported to inhibit corneal neovascularization, showed its clinical application, however, Avastin treatment of corneal neovascularization optimal concentration has not been determined. The experimental observation of three concentrations of Avastin in rat corneal alkali burn-induced neovascularization for clinical dosing regimens provide experimental basis. The first part of the different concentrations of Avastin rat corneal neovascularization Objective To observe the dynamics of different concentrations of alkali burns avastin induced corneal neovascularization, and explore the optimal drug concentration avastin. Methods prepared by alkali burn rat corneal neovascularization. Modeling were randomly divided into control group, experimental group 1, group 2 and Experiment 3 experimental groups of 15 rats. Control group: alkali burn and burn immediately after 7d in the subconjunctival injection of 0.05ml saline. Experimental Group 1: subconjunctival injection avastin 0.05ml (2.5mg/ml); experimental group 2: subconjunctival injection avastin 0.05ml (12.5mg/ml); experimental group 3: subconjunctival injection avastin 0.05ml (25mg / m1); each experimental group were alkali burns and the burn immediately after 7d subconjunctival injection of different concentrations corresponding equivalent avastin. Observed the growth of corneal neovascularization after alkali burn and compare 7d, 14d, 28d each group the difference between the area of ??corneal neovascularization. Alkali burns after 7d, 14d, 28d, respectively, in each group to take five rat cornea HE staining, light microscope. Results of the first 7d, 14d, 28d each experimental group corneal neovascularization area were significantly less than the control group, the differences were statistically significant (P lt; 0.05), experimental group 2, the experimental group were less than three experimental group, the differences were statistically significant (P lt; 0.05); experimental group 2, experimental group 3 CNV area at different points in time no significant difference (P gt; 0.05). Conclusion 2.5mg/ml, 12.5 mg / ml, 25 mg / ml in three concentrations to 12.5mg/ml avastin alkali burn corneal neovascularization inhibition better, avastin subconjunctival injection was not found after systemic and local side effects. Experimental Group 1: subconjunctival injection avastin 0.05ml (2.5mg/ml); experimental group 2: subconjunctival injection avastin 0.05ml (12.5mg/ml); experimental group 3: subconjunctival injection avastin 0.05ml (25mg / ml); each experimental group were alkali burns and the burn immediately after 7d subconjunctival injection of the same amount corresponding concentrations avastin. Alkali burns after 7d, 14d, 28d, respectively, was observed in each group after taking five rat cornea, immunohistochemical detection of the expression of VEGF. Compare alkali burns after 7d, 14d, 28d each group of VEGF positive expression rate differences. Results after corneal alkali burn 7d, 14d, 28 d, experimental group and control group 1,2,3 corneal tissue expression of VEGF positive rate there was a significant difference between the control group, the positive expression rate is higher (P lt; 0.05) each time point experiment a group of high VEGF expression in Experiments 2 and experimental group 3 (P lt; 0.05), while the experimental group 2 and VEGF expression in experimental group 3 was no significant difference (P gt; 0.05) Conclusion alkali burns corneal neovascularization and VEGF are closely related, avastin corneal alkali burn VEGF expression has an inhibitory effect. 2.5mg/ml, 12.5mg/ml, 25 mg / ml to 12.5mg/mlavastin three concentrations of VEGF inhibition of corneal alkali burn better.
