Objective: dendritic cell chemokine produced by dendritic cells (dendritic cells, DCs) 1 (dendritic cell-chemokinel, DC-CK1) play a key role in the start of the initial immune response . Leukemia source DCs observed in vitro induced expression levels of DC-CK1 , to find new ways for the development of leukemia immunotherapy . Method : acquisition of newly diagnosed acute myeloid leukemia ( AML ) , acute lymphoblastic leukemia ( AALL ) , chronic myeloid leukemia (CML) in peripheral blood of patients and healthy volunteers , with lymphocyte separation medium density gradient centrifugation mononuclear cells (monouclear cells , , PBMC), with recombinant human granulocyte - macrophage nuclear cells set off the stimulus factor ( rhGM - CSF ) , 100 ng / ml , recombinant human interleukin 4 ( of rhIL - 4 ) 100 ng / ml , and tumor necrosis factor- a ( of TNF - α ) 50 ng / ml joint training of 8 days, cell morphology ( inverted microscope , to identify DCs cultured Wright staining ) and phenotype ( flow cytometry ) ; fluorescence in situ hybridization (FISH) cytogenetic DCs learn detection ; semi-quantitative PCR (RT-PCR) to detect the DCs expressed the DC-CK1mRNA level ; enzyme-linked immunosorbent assay (ELISA) detected in the culture supernatant concentration of DC-CK1 . Results: the Leukemia source DCs and normal DCs morphological changes similar to their surface marker expression there is no obvious difference between the positive rate with normal DCs (P gt ; 0.05) . RT-PCR and ELISA showed that AML- DCs CML -DCs and ALL-DsC , , the expression of the DC-CK.1mRNA and levels of DC-CK1 were 0.26 ± 0.025 ( 152.32 34.74 ) pg / ml , 0.27 ± 0.020 ( 163.06 ± 40.97 ) pg / ml and 0.29 ± 0.034 ( 173.80 ± 40.44 ) pg / ml , significantly lower than the normal -DCs 0.43 ± 0.032 ( 325.36 ± 39.23 ) pg / ml, and the difference was statistically significant (P lt; 0.05). Conclusion: The results suggest that : the mRNA and protein levels in a variety of cytokines induced leukemia source DCs its DC- CK1 trend , were significantly lower than the normal of DCs Leukemia source DCs to induce an immune response which indicates reduced ability . By raising the level of expression of DC-CK1 , may help to strengthen the combination between leukemia source of DCs with initial lymphocyte beneficial to initiate an immune response , in order to enhance the anti-tumor effect of leukemia source DCs , clear minimal residual disease and reduce relapse of leukemia has a very important significance .
