Abstract


Phosphorylated glycoprotein extracellular matrix / osteoblast / bone cells factor 45 (Matrix Extracellular PhosphoglycoprotEin / Osteoblast / Osteocyte Factor 45, MEPE/OF45) was first cloned in 2000 since the N-terminus as a small connection integrin glycoprotein (Small Integrin-Binding Ligand, N-linked Glycoprotein, SIBLING) family of which the function has been limited research in the regulation of bone metabolism and phosphorus (in vivo ) balance . In recent years, our laboratory and foreign collaborating laboratory preliminary study reveals protein MEPE/OF45 new features , namely MEPE/OF45 involved in cellular DNA damage response ; MEPE/OF45 can enhance inducer cells to DNA damage (DNA Damage Inducers ) tolerance. Our laboratory studies using DNA microarray MEPE/OF45 functional process also found MEPE/OF45 can inhibit apoptosis, and accompanied by high expression MEPE/OF45 SERPINB2 [Serine (or cysteine) proteinase inhibitor, clade type-2 ] and SERPINB2 related protein RB (retinoblastoma) high expression . Therefore, the study of the regulation of relations between the three possible to reveal the MEPE/OF45 anti-radiation and anti- apoptotic function is important. In the present study, we obtained the following results: ( 1 ) the SerpinB2 gene prokaryotic expression and purification of fusion protein SERPINB2 and MEPE/OF45 and mice immunized with the purified protein preparation and the anti SERPINB2 polyclonal anti MEPE/OF45 antibody, the antibody was measured by ELISA titer 1:20000, Western blotting showed that the antibody specificity for ; ( 2 ) SerpinB2-siRNA vector was constructed , screened two SerpinB2-siRNA, 293T cells can be endogenous to inhibited the expression of SERPINB2 ; ( 3) transient expression of transfected MEPE/OF45, microRNA expression and inhibition MEPE/OF45 SERPINB2 siRNA knock down the expression of the MEPE/OF45 SERPINB2 and RB with the relationship between the protein level found MEPE/OF45, SERPINB2, RB protein levels are consistent , it may be one of the ways MEPE/OF45 affect apoptosis . The work will not only help deepen understanding of cellular DNA damage response mechanism for understanding radiation therapy may also target molecule for future research to provide meaningful clues.
