Abstract

The brown ring glued cap boletus is an excellent ectomycorrhizal and has nutritional value and medicinal value . Bacteria in the culture medium after a certain time , the mycelial biomass with the initial pH change , two peaks, and when the initial pH is higher than 5.5 , the reduction of the pH of the liquid medium, large amplitude . In order to investigate the reasons for the phenomenon , and to understand the impact of the initial pH on mycelial growth and medium acidification , the experiments so brown ring sticky cover on the of Boletus growth medium at different initial pH conditions through mycelial Determination of biomass , medium pH indicators brown ring the sticky cap boletus mycelium with medium pH interaction . The results show that the mycelium Exudation of type and amount vary under different initial pH , of pH4.5 ~~ 7.0 under conditions will produce oxalic acid , pH5.0 under conditions also produce acetic acid . With the initial pH increased , the amount of the unit mycelium secretion of oxalic acid is increased. Increasing share of the total H to increase the percentage of the concentration of the culture medium with the pH increases the concentration of oxalic acid ionization of H in the range of pH4.5 ~ 6.0 , at pH 7.0 oxalate ionized H concentration of the culture medium increased H a concentration of 16 times. Ability pH6.0 under the conditions of the pre - cultured hyphae secretion of oxalic acid . With prolonged incubation time , the oxalic acid secreted H culture acidification weakened. Significant correlation between the changes in the pH of the medium and reducing sugar consumption and mycelium biomass . Culture the 12d since pH4.5 ~ 7.0 under the conditions of oxalic acid secretion rate slows down. Under the conditions of initial pH6.0 , a potassium ion on the mycelial growth inhibition, with the potassium concentration increased , the H secretion rate decreased significantly . Medium pH conditions will affect the metabolism of organic acids mycelial pathway, whereas mycelium also secrete certain substances causing changes in the pH of the medium .
