Abstract


Objective To detect NRP1 expression in human gastric cancer ; design NRP1 small interfering RNA plasmid ; neuropilin 1 (neuropilin 1 NRP1) implanted gastric tumor growth inhibition by RNA interference technology to explore . By immunohistochemistry SP method determination NRP1 expression in human gastric cancer and NRP1 expression in normal gastric tissue , and microvessel density was measured in the number of vascular endothelial cells with CD34 marker ; design hairpin siRNA targeting human NRP1 gene ( Small interfering RNA SiRNA) sequence , the synthesis of a complementary oligonucleotide annealed and cloned to the plasmid pGenesil - 1.1 recombinant plasmid by restriction analysis and DNA sequencing to build ; human gastric cancer cells SGC7901 injected into nude mice right buttocks subcutaneous xenograft model ; recombinant plasmid was injected into the tumor to nude mice and nude mice in general and the determination of the weight and volume of nude mice ; pathological observation of tumor tissue and blood vessel growth ; immunohistochemical methods observed nude mice in to NRP1 the expression and microvascular density . NRP1 expression and microvessel density in human gastric cancer was significantly higher than that in normal gastric tissue was statistically significant (P lt; 0.05); restriction analysis and DNA sequencing confirmed that the purpose of oligonucleotide fragments were cloned into plasmid pGenesil-1.1 ; the weight and volume of the xenografts in nude mice in the experimental group than the control group ( P lt; 0.05 ) ; biopsy shows the experimental group tumor necrosis were markedly ; immunohistochemical tumor cells in nude mice in the experimental group NRP1 expression and microvessel density lower than the control group ( P lt; 0.05 ) . NRP1 expression in the conclusions of gastric cancer than normal tissue increased NRP1 expression and microvessel density associated with increased ; through the the SiRNA-NRP1 inhibition of NRP1 expression implanted gastric cancer , can affect angiogenesis in nude mice , and ultimately inhibit tumor growth.
