Abstract


Objective: To investigate curcumin curcumin for clinical treatment of colon cancer in nude mice vincristine resistant human colon cancer cell lines HCT-8/VCR multidrug resistance reversal effect and its possible mechanism , to provide a theoretical basis . Method : HCT-8/VCR cells were grown in nude mice subcutaneous tumor resistance model . 20 nude mice were randomly divided into four groups : group A : control group ; B group : VCR group ; C Group: curcumin group ; D group : Curcumin Joint VCR group . Every the 1d measuring tumor longest diameter and shortest diameter and calculate its volume ; collect specimens , said two weeks after the tumor weight ; the inside of MDR1 mRNA in tumor tissue were detected by RT-PCR and Westernblot , of Survivin mRNA and P-gp, Survivin protein expression. Preparation tumor histopathology observed tumor tissue cell morphological changes . Results: Curcumin combined with VCR significant inhibition of tumor growth and tumor weight was significantly lower than the rest of the group ( P <0.05 ) . RT-PCR, Westernblot results show that curcumin joint VCR group and the curcumin group , the MDR1 mRNA , SurvivinmRNA and P -gp , survivin protein expression significantly decreased compared with the control group , the difference was statistically significant ( P <0.05 ) . The biopsy revealed more lymphocyte infiltration curcumin group and curcumin VCR group compared with the control group and the VCR group the karyopyknosis more mitotic , more extensive necrosis . Conclusion : Curcumin can reverse colon cancer multidrug resistance , may become colon cancer multidrug resistance reversal agents .
