Objective: To evaluate astrocyte glutamate - glutamine cycle in rats with neuropathic pain . Methods: 48 adult male SD rats , weighing 200 ~ 230 g, were randomly divided into 8 groups ( n = 6 ) : control group (C ) does not carry out any processing ; sham surgery group (S ) is not only exposed nerve ligation or transection ; neuropathic pain group (NP group ) using ligation and transection model prepared tibial nerve and common peroneal nerve ; models were successfully after the NP group intrathecal injection of PBS 50 μl of the experimental group (M1 ~ 4 groups ) intrathecal injection the 0.01,0.03,0.05,0.10 mmol / L glutamine synthetase inhibitor MSO 50ul, MSO Gln group (MG group ) intrathecal injection of 0.05 mmol / L MSO 50 μl of 15 min after the intrathecal injection of 0.25 mmol / L glutamine 50μ1. In one week prior to ligation (TO), one week after ligation (T1), after injection of MSO 15,30,45,60 min (T2 ~ 5) determination of the mechanical pain threshold , rats were killed after the last time measured pain threshold L4 ~ 6 segment of the spinal cord , using immunohistochemistry staining of glial fibrillary acidic protein (GFAP) and glutamine synthetase (GS) expression and co-expression (GFAP / GS) . Results: Compared with the S and C groups , NP group and the MG group T1-5 mechanical threshold reduced; compared with the NP and the MG group M3 group T2-4 mechanical pain threshold increased , T5 when the MG group and NP spinal cord dorsal horn GFAP, GS and GFAP / GS , upregulated ( P lt; 0.05 ) ; these indicators MG group and NP group difference was not statistically significance ( P gt ; 0.05) . Conclusion: the astrocyte glutamate - glutamine cycle involved in neuropathic pain .
