Abstract


Objective To exploring the pathogenesis of the chronic hepatitis B (CHB).Method 1. The distribution of HBcAg in liver cell of patients was analyzed by the method of enzyme linked immuno-histochemistry. 2. Pathological diagnosis was performed using liver biopsy for assay the HBcAg expression of localization in hepatocytes by an immuno-fluorescence confocal technique; 3. The method for separating cytoplasm and membrane protein form liver cells was by the protein extraction kit; 4.To characterize the HBcAg compounds with western blot lexes.Results By enzyme-linked immuno-histochemistry and microscope, in 41 patients with liver biopsy, it has found positive expression of HBcAg in 24 cases, and in the same patient HBcAg may appear different expressive patterns, mainly of cytoplasm and nucleus type, but often in a unique pattern. Including colony, diffuse, or irregular of distribution, the weaker of HBcAg expression, the more irregular of distribution. The results of confocal microscope observation showed that positive rate of HBcAg expression in liver cells was 87.8% in 36 patients, in which there were 23 cases with abnormal liver function, 10 with mild abnormal liver function, 3 with normal liver function. HBcAg expression showed the cytoplasm-type and nuclear-type, no membrane-type ones. The liver injury is obvious connected the patterns of membrane-type expression of HBcAg in liver cells. It also showed that mixed cytoplasmic and membrane expression of normal liver function only in a small group found karyotype expression. In addition, by Western blot method, cytoplasm and membrane protein of HBcAg was detected in liver cell of patients with unnormal liver function.Conclusions Hepatitis B virus in patients with chronic infection may cause pathological changes in liver, the immunological damage of liver cells are resulted from the advents of membrane expression HBcAg, and it is to prove that the HBcAg expressed on cell membrane is the target antigen of immune response in chronic hepatitis B (CHB) patients.
