ObjectiveTo detecet the expression of X-1 inked inhibitor of apoptosis protein (XIAP) and Second mitochondria-derived activator of caspase (Smac) in normal cervical tissue, cervical atypical hyperplasia (CIN) and cervical squamous cell carcinoma, and discuss their significance in the occurrence and development of cervical squamous cell carcinoma.MethodsCollecting 125 samples of patients with complete clinical data and pathologically confirmed, including 60 cases of cervical squamous cell carcinoma,45 CIN cases and 20 normal cervical cases. To detect the expression differences of XIAP and Smac by Immunohistochemisry SP in different cervical tissueResults1. XIAP expression in different tissues of the cervixIn normal cervix, CIN and cervical squamous cell carcinoma group, the positive expression rate of XIAP separately is 10%(2/20),40% (18/45),78.3%(47/60), the overall rate of positive expression shows significant difference (P<0.05). Pairwise comparison among the three groups, the difference is statistically significant (P<0.017). There is no significant difference when CINI is compared with CINII (P>0.017), and also between CINII and CINIII (P>0.017), but the difference between CINI and CINIII is significantly(P<0.017)。In cervical squamous cell carcinoma group, the expression of XIAP is associated with cell differentiation, and lower the degradation of the tissues is, XIAP expression higher. The expression shows significant difference in high, medium and low differentiation groups (P<0.05), and there is significant difference in expression between well-differentiated and mid-differentiation groups, and also between well-differentiated and poorly-differentiated groups (P<0.017), but between mid-differentiation and poorly -differentiated groups, there is no statistically significant difference (P> 0.017). XIAP expression is unrelated to age, clinical stage (P>0.05).2. Smac expression in different tissues of the cervixIn normal cervix, CIN and cervical squamous cell carcinoma group, the positive expression rates of Smac separately is 95%(19/20),64.4% (29/45),31.7%(19/60), the overall rate of positive expression shows significant difference (P<0.05). Pairwise comparison among the three groups, the difference is significant (P<0.017). There is no significant difference when CINⅠis compared with CINⅡ(P>0.017), and also between CINⅡand CINⅢ(P>0.017), but the difference between CINI and CINⅢis significantly(P<0.017)。In cervical squamous cell carcinoma group, the expression of Smac is associated with cell differentiation, and lower the degradation of the tissues is, Smac expression lower. The expression shows significant difference in high, medium and low differentiation groups (P<0.05), and high differentiation and the differentiation of well-differentiated and poorly differentiated group and between group differences in the expression of both (P<0.017), but the poorly differentiated and there was no significant between groups was statistically significant (P>0.017). Smac expression is unrelated to age, clinical stage (P>0.05).3. The correlation of XIAP and Smac expressionIn cervical squamous cell carcinoma tissue,XIAP and Smac expression shows negative correlation ,and correlation coefficient r=-0.599 p<0.05.Conclusion1. In normal cervical tissue, CIN tissues and cervical carcinoma, the XIAP expression level increases gradually, and expression in cervical carcinoma is significantly higher than normal cervical tissue. It suggests increasing expression of XIAP may be related to the occurrence of cervical squamous cell carcinoma tissue.2. In normal uterine cervix, CIN tissues, cervical carcinoma, the Smac expression level decreases gradually, and expression in cervical carcinoma is significantly lower than normal cervical tissue. It indicates decreasing expression of Smac may be related to the occurrence of cervical squamous cell carcinoma tissue.3. XIAP expression in cervical squamous cell carcinoma increases, while Smac expression decreases. The expression of the two is negatively correlated. The interaction between them may promote the occurrence of cervical squamous cell carcinoma tissue.
