Abstract


ObjectiveThe previous preparation of chemically extracted acellular nerve allograft were based on rats, rabbits, dogs and other animals. Based on the previous and existing literature,we selected the sciatic nerve of pig as the object of study. The chemically extracted acellular nerve to pig’s sciatic nerve are prepared by different experimental methods and different reagents .we studied on the chemical extraction to the larger nerve and compared the degrees of decellularization of different methods then we could choose the better methods for chemical extraction of larger nerve.MethodThe pigs were exposed by a muscle-splitting incision .The fat and fascia on the sciatic nerve membrane were removed. A total of 30 segments of sciatic nerve were taken. Each was 60 mm in length and 4mm in diameter. The segments were divided into 3 groups by different methods of chemical extracting.GroupⅠ: the Sondell method group, in which the nerve segments were chemically extracted with the detergents of Triton X-100 and deoxycholate for two times.GroupⅡ:The chemical detergent-enzyme digestion method group in which the nerve segments were chemically extracted with sodium dodecyl sulfate,Deoxyribonuclease I,Ribonuclease I and Ethylene diamine tetraacetic acid for 48 hours .Control group : untreated sciatic nerves of pig . The degrees of integrity of nerve fiber tube and demyelination were observed under microscope and were assessed by a scoring system.ResultsIn both groups the integrity of nerve fiber tube was preserved well but they were not better than the control group .The decellularization of groupⅡwas better than groupⅠ. In groupⅡ, more laminin component was saved and the damage to allograft nerve was smaller. In the preparation of chemically extracted acellular nerve allograft, we should consider the saving of laminin component ,the degree of decellularization and the integrity of nerve fiber tube.Conclusion1 In the study of the methods of chemically decellularization to the sciatic nerve of pigs , we found the chemical detergent - enzyme digestion method may be an ideal choice for the preparation of chemically extracted acellular nerve allograft for larger nerve cells. With this method, we can preserve the structure of the nerve better and remove the immune antigens more thoroughly, Such method has good prospects for development.2 Chemically extracted acellular allograft can significantly promote the the neurotropic regeneration.3 Chemically extracted acellular allograft nerve is an ideal graft subsitutes, it acted as a scaffold in the period of large segements of nerve defects repair and markedly enhance the effects of neurotropic reinervation, which artificial synthetic material can not .we believe that with the development of technology, the chemically extracted acellular allograft may provide an alternate to the autologous nerve transplantation.
