Association of A1166C Polymorphism in AT1R Gene with the Occurrence of AMI and the Extent of Coronary Artery Stenosis in AMI Patients

Objective: To study the relationship between A1166C polymorphism in angiotensinⅡtype 1 receptor (AT1R) gene and occurrence of acute myocardial infarction (AMI) and the extent of coronary artery stenosis in AMI patients. According to the results, we may find a new theory to prevent or treat AMI patients on the basis of their medical conditions.Methods: The A1166C polymorphism of 105 primary AMI patients and 111 healthy control subjects was detected by polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP). All AMI patients underwent coronary angiography and the number of affected coronary vessels (≥50% stenosis) or the Gensini score were determined by the results of coronary angiography. Then some risk factors of coronary artery disease(CAD) were assessed. The relationship between A1166C polymorphism in AT1R gene and occurrence of AMI, the number of affected coronary vessels (≥50% stenosis) and the Gensini score in AMI patients were explored.Results: Neither the genotype frequencies of AA, AC + CC (P>0.05) nor the allele frequencies of A and C (P>0.05) differ significantly between the patients and controls. The genetic and environmental risk factors were studied by Logistic regression analysis. The results showed that hypertension, family history of CAD, high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C) were closely related to AMI(P<0.05). HDL-C was the protective factor (OR<1, 95% CI 0.155~0.931), the remaining three were risk factors (OR>1). Either the number of affected coronary vessels or the Gensini score in AC+CC genotype carriers was significantly higher than that in AA genotype carriers(P<0.05).Conclusions: The A1166C polymorphism in AT1R gene is not associated with the occurrence of AMI but the extent of coronary artery stenosis(number of affected coronary vessels and Gensini score).
