Abstract

Objective Huang oligosaccharides capsules for quality research and develop quality standards. On this basis, it was observed in diabetic rats spatial learning and memory. Method in accordance with the chosen technology of Rehmannia glutinosa oligosaccharides capsule preparation, refer to \Mainly on the formulation ingredients contained stachyose were identified by thin layer chromatography and high performance liquid chromatography using (HPLC) determination of which catalpol content. On this basis, further observation of Rehmannia glutinosa oligosaccharides (ROS) in diabetic rats spatial learning and memory in rats. Prepared using streptozotocin diabetic rats were divided into control group (CON), normal treatment group (ROS, 500 mg / kg), diabetes mellitus (DM), oral administration of 100 mg / kg to low ROS diabetes treatment dose group (DM RL) and oral 500 mg / kg to yellow oligosaccharides diabetes high dose group (DM RH). Regular monitoring of animal body weight during the experiment, and fasting blood glucose values. 11 weeks after the Morris water maze learning and memory abilities of rats changes. Results using the TLC Huang oligosaccharides capsules stachyose were identified, the test results are displayed in the chromatogram chromatography with reference substance corresponding position, with the same color spots. Ground yellow oligosaccharides in catalpol content were measured, the results show that catalpol detection method in the range of 10 ~ 90μg/mL showed a good linear relationship, Y = 3.085 × 10 ~ 3X-2.060 × 10 ~ 3, r = 0.9996; RSD of 0.35%; repeatability RSD was 1.09%; stability RSD was 1.02%; recovery was 99.84%, RSD was 0.95%. Take the lot number 20090501,20090503 and 20090506 Huang oligosaccharides capsules, as measured by HPLC in which the content of 12.12,12 catalpol 56 and 12.37 mg / tablets. Huang oligosaccharides capsule disintegration load difference ﹑ moisture content are in line with the \Huang oligosaccharides Capsule on learning and memory in diabetic rats found: diabetes (DM) group ﹑ low-dose treatment group (DM RL) and high-dose treatment group (DM RL) and the control group (CON) group, significant weight reduce (P lt; 0.01); blood glucose increased significantly (P lt; 0.01); Morris water maze experiment, the three groups than the control group in the target quadrant time the proportion of the total swimming time was significantly reduced, while the diabetic group than the control group was significantly longer escape latencies; and DM group, DM RL group and DM RH group escape latency was significantly shorter in the former platform quadrant swimming time total swimming time was significantly increased; medication alone group (ROS) and the control group (CON) group, no significant change. Conclusion 1 ﹑ established method of identifying specific, quantitative method is simple, accurate, and can effectively control the quality of Rehmannia glutinosa oligosaccharides capsules. 2 ﹑ diabetic rats spatial learning and memory decline, to yellow oligosaccharides capsule on improving the ability of learning and memory in rats with diabetes have some therapeutic effect.
