Research purposes from the antioxidant enzymes, histology, and ultrastructure three levels to reveal the impact of 20 weeks of regular aerobic exercise on middle-aged rats arterial wall structure and anti-oxidative capacity, at the same time for 20 weeks of regular aerobic Retard the vascular effects assessed. 2 research methods 30 18-month-old healthy female Sprague-Dawley (SD) rats, Hunan Agricultural University experiment Faculty. Were randomly divided into 2 groups: a control group of middle-aged, middle-aged the incremental intensity aerobic exercise group, n = 15; another: young control group, 6-month-old young SD rats, a total of 15. National Laboratory Animal husbandry standards were housed separately. Treadmill movement, the application of blood lactate indicators monitoring exercise intensity, the development of middle-aged rats incremental load intensity aerobic exercise program. Lactic threshold test program: cut tail blood acid --- the power curve for the principle of measured the reference of Behavioral Medical lactate threshold determination treadmill incremental load method. Initial load of 15m/min, the duration is 2-3min, increment in 3m/min, intermittent time was 30s. Rat exercise program: Phase I: adaptation to the environment, not training; Phase II: adapt to treadmill training; Third stage: the increasing load aerobic exercise daily to the required strength. As follows: the first week of the speed of 15m/min movement, each time for 15min; follow the single-week increase speed Fortnight increase the time the principle of turn increments, the incremental time range: 5min/week; incremental velocity amplitude 1m/min/week. Training six days a week, Sunday. A total of 20 weeks of exercise. After the expiration of the sport, 12 hours after the end of the exercise program, weighed and recorded, with 0.4% of chloral hydrate lml/100g weight anesthesia, the rats were supine fixed in anatomy table, orbital blood, anatomy of the rat exposed chest, the blood drawn by low temperature centrifuge at 4 ℃, 2500r/min centrifuge for 10 minutes, do not have the centrifugal a good blood specimens from the supernatant was discarded following precipitation. Detection of the blood after take the rat thoracic aorta segment for advanced oxidation protein products (Aopps) detection and morphology. Spectrophotometer SOD, NO and GSH-Px activity detection, use a double antibody two-step sandwich enzyme-linked immunosorbent assay (Elisa) to detect the content of Aopps, by means of the arterial wall by light microscopy and electron microscopy Histologic and ultrastructural detection. 3. 3.1 blood lactate test results of the findings from the quiet state to exercise intensity 24m/min when blood lactate values ??change slightly, at a relatively constant change. When the speed is greater than 24m/mim, blood lactate values ??increased, indicating glycolysis. Therefore, the the rat exercise intensity is not higher than 24m/min. The 3.2 regular aerobic exercise on antioxidant enzymes in rats detection and advanced oxidation protein products (1) elderly control group and the exercise group of middle-aged artery homogenate Aopps content higher than the younger control group, and has a very significant difference (P lt ; 0.01), middle-aged the exercise group Aopps content lower than the control group of middle-aged, and has a very significant difference (P lt; 0.01) (2) middle-aged exercise group and the control group of middle-aged rat plasma SOD activity was lower than younger the control group, the control group of middle-aged rats and young rats in the control group, SOD activity has very significant difference (P lt; 0.01); SOD activity of the exercise group of middle-aged rats with middle-aged control rats, with a significant difference (P lt; 005) (3) exercise group of middle-aged and middle-aged control rats plasma GSH-Px activity were lower than the younger control group, the control group of middle-aged rats and young control rats GSH-Px activity has very significant difference (P lt; 0.01); higher than the control of the middle-aged, middle-aged exercise group, GSH-Px activity and exercise group of middle-aged and the middle-aged control rats GSH-Px activity with a significant difference (P lt; 0.05). (4) the exercise group of middle-aged and middle-aged control group, plasma NO levels were lower than the young control group, the control group of middle-aged and young control group, NO content has a very significant difference (P lt; 0.01), the exercise group of middle-aged and NO content of the younger control group with significant difference (P lt; 0.05); NO content of middle-aged exercise group than the control group of middle-aged, and has a very significant difference (P lt; 0.01). 3.3 rat aortic HE staining results compared with young control group, the control group of middle-aged inner diameter (d), the diameter (D) and arterial wall thickness (ADT) showed a very significantly increased (P lt; 0.01), middle-aged exercise group d and D was significant resistance increase (P lt; 0.05), ADT was very noticeable with sexual increase (P lt; 0.01); compared with the middle-aged control group, middle-aged sports group d, D and ADT was very noticeable with nature to reduce (P lt; 0.01) 3.4 rat vascular elastic fibers the EVG stain results young control rats elastic fiber membrane deeply stained uniform, the structural integrity of the film, wavy folded rich, folding a large range, and even showed a \aged control rats, lighter and thinner membrane staining of elastic fibers, and even a fracture, only very sparse folded, and fold the number and magnitude is very small; deeper exercise group of middle-aged rats membrane staining of elastic fibers, wavy folding the number and magnitude of the increase; 3.5 rat aortic collagen fibers immunohistochemical staining results compare with the younger control group, positive control group of middle-aged expression was significantly increased (P lt; 0.01), the positive expression of middle-aged exercise group was significantly increased (P lt; 0.05); compared with the control group of middle-aged, middle-aged exercise group positive expression was a significant reduction (P lt; 0.01). The young control group of 3.6 the rat aortic electron microscopy results you do not see any of endovascular aortic abnormalities; control group of middle-aged local the slurry bubble necrosis formation of vacuoles, serious at was mucus viscous, elastic fiber breakage of the entire wall, collagen fibers increased significantly and disordered arrangement; mild exercise group of middle-aged endothelial cell damage, elastic fibers are arranged more regularly. Conclusion 4.1 20 weeks of regular aerobic exercise can improve in old rats antioxidant enzyme activity; 4.2 20 weeks of regular aerobic exercise can change the arterial wall changes associated with aging, slow down the aging process of the arterial wall structure; 4.3 by 20 weeks of regular aerobic exercise, reduce Aopps accumulation induced oxidative stress and stimulation, and thus achieve the improvement of oxidative stress on endothelial cells.
