Considering the main problems existed in the present products of cataplasm on sale, for example, the viscidity of cataplasm is deficient, and the cataplasm is apt to left over when it is heated, we optimize prescription of Cataplasm which is composed of sodium polyacrylate(B) and polyacrylate (A) as the skeleton material on the basis of model drug-sinomenium acutum in this experiment. By comparing their performance, we select the better type of Cataplasm, optimize their preparation process, and conduct three experiments as vitro release, pharmacokinetic and toxicology. Finally, we draw the following conclusions:1. Taking the beginning of tack and tack holders as the index, we acquired the optimized prescription of type A and B respectively through single factor and orthogonal tests. It is found cataplasm B is better than A obviously and its best prescription is: 5.6% of the part of sodium polyacrylate, 38% of Glycerol, 0.28% of Glycine aluminum and equivalent Tartaric acid.2. Taking the beginning of tack and tack holders as the index, we obtain the best conditions for production of Cataplasm B, that is, disperse the drug and the part of sodium polyacrylate in glycerol simultaneously, agitate for 30 minutes, and then put it under the temperature of 45℃.3. By investigating the effect of cataplasm B after different oil phases being added on the model drug in vitro release, we draw a conclusion that the effect of percutaneous absorption is better than the simple hydrophilic cataplasm when castro oil is used as the oil phase, and its permeation amount is 2.8 times as that of hydrophilic cataplasm.4. By studying the effect of cataplasm B after different types of penetration agents being added on the model drug in vitro release, we find that the effects of different types of penetration agents are as follow: cineole> propylene glycol> Tween> Azone, and the maximum amount is 1.66 times as the minimum.5. The pharmacokinetic study on the percutaneous absorption of sinomenium acutum cataplasm and the oral administration of sinomenium acutum containing castor oil and eucalyptus oil, it is showed that the Tmax of the cataplasm is 12 hours, which prolongs the peak time compared with the oral administration (2 hours), and that the Tmax of the cataplasm is 1.537μg/ml, which decreases compared with the oral administration (6.521μg/mL).
