Effect of Esculentoside a on Apoptosis and Expression of PCNA、Caspase-3、Fas and FasL in Renal Tissue of BXSB Mice

Objective:To explain the mechanism of EsA effects LN and effects on the expression of PCNA、Caspase-3、Fas and FasL in renal tissue of BXSB mice.Methods:24 male BXSB mice at the age of 18 weeks were randomly divided into three groups:model group、EsA-treated group and Dexamethasone-treated group. Each group were treated,all mice were obtained urine randomly,and were killed to obtain renal tissue after 4 weeks. The following parameter was detected:1.Urine protein and urine creatine concentration;2.HE staining of renal pathology.3.The expression of PCNA and Caspase-3 were detected by in stiu hybridization in renal tissue;4.The expression of Fas and FasL proteins were detected by immunohistochemtry in renal tissue.Results:1.At last,the level of urine protein/urine creatinine was decreased in EsA-treated group and Dexamethasone-treated group (P<0.05);There is no difference between EsA-treated and Dexamethasone-treated group(p>0.05);2.After using drug,the renal pathological change of BXSB mice moderates significantly;3.The expression of PCNA mRNA in renal tissue of model group was significantly higher than it of EsA-treated group and Dexamethasone-treated group (P<0.05).There is no diference between EsA-treated group and Dexamethasone-treated group(P>0.05);4.As for,the expression of Caspase-3 mRNA、Fas and FasL in renal tissue of model group was lower than them of EsA-treated group and Dexamethasone-treated group,and there has significance in three groups(P<0.05),but there is no difference between EsA-treated group and Dexametha-sone-treated group(p>0.05).5.Correlation showed that there was no correlation between the expression of Caspase-3 and Fas in mesangial cell in three groups.Conclusion:1.Experimental doses of EsA and Dexamethasone can degrade urine protein concentration and improve renal pathology,so they can reduce renal damage of BXSB mice;2.EsA and Dexamethasone can reduce renal damage by the potenial meshanism of inhibiting renal cell proliferation and improving renal cell apoptosis of BXSB mice.3.There was no correlation between the expression of Caspase-3 and Fas of mesangial cel of BXSB mice each group.
