Infectious diseases is one disease of the clinic diseases, which are caused by bacterium virus mycoplasma Chlamydia richettsia fungal parasite etc. The bacterial infectious diseases occupy sizable percentage among these clinic diseases. Before the discovery of the antibiotics, this type diseases harmed for the human’s life and healthy. The occurance and application of antibiotics have educed enormous effects on preventing and treating bacterial infectious diseases. But with the unstandard using of antibiotics, many serious problem have occurred, such as microorganism drug-raistance bacteria , antibiotics accumulate adverse reaction and the passive influence for the immunity function. Traditional Mongolian medicine and pharmaceutical science is the most important compose of Chinese medicine treasury. It has significant curative effect on bacterial infectous diseases in traditional Mongolian medicine prescription dose. Reseach has indicated, In the mechanism of anti-infection, some medicine can treat infectious diseases through direct repressing the bacterium or regulating the immunity function to strengthen the organism’s immunity to transfer self-potential to achieve the purpose of treating the disease. The Mongolian medicine SanZi Decoction is composed with myrobalan、gardeniae and toosendan. Amounts of experiement have proved that the single medicine of this prescription dose has the capability of broad-spectrum antibiosis, but there is no reports of researching on SanZi Decoction to repress bacterium and regulate the immunity function. This experiement apply SanZi Decoction to observe its effects of antibiosis in vivo and regulating the immunity function, providing the basis of using medicine in clinic.Purpose:Research the effect of antibiosis of SanZi Decoction and its regulating function on immunity.Method:We use artificial bacterial infection animal model to research the antibiosis and observe the protection of SanZi Decoction to the infectious animals. We use vivoand vitro ways to study the regulating immunity function of SanZi Decoction. The vivo ways: we give SanZi Decoction in vivo, to observe the effect of the medicine on immunity organs weight of animals、the effect of spleen-lymphocytes proliferation induced by ConA and the effect of phagocytize capability of macrophage in abdominal cavity. Vitro ways: observe the effect of SanZi Decoction on spleen-lymphocytes proliferation and the ph-agocytize capability of macrophage in abdominal cavity culture in vitro .Result:1.The Sanzi Decoction have protective effect on mice infected with Staphylococcus or P.Aeruginosa. Significantly brought down the mortality rates of infected mice . 2.The different dose groups’thymus index and spleen index are both higher than the control group slightly, But after statistics analysis, the difference is not significant (P>0.05).3.The phagocytosis rate and phagocytic index of three different dose group are all higher than control group, especially the middle dose group(P<0.01). 4. Three different dose groups’absorbance values were higher than control group slightly, but no significant difference were found between them (P>0.05).5.The different concentration of Sanzi Decoction (100ug/ml, 50 ug/ml,25 ug/ml,12.5 ug/ml) can directly stimulate spleen lymp- hocyte proliferate in vitro, degree of cell prolife-ration increased with the dose of Sanzi Decoction.6. Except the concentration 12.5 ug/ml group, the other groups all can promote ConA induced spleen lymphocyte cell proli-feration in vitro, The results showed that the proliferation was increased by Sanzi Decoction in concentration-dependent manner. 7.Except the concentration 12.5 ug/ml group, the other groups enhance the phagocytic function of the mice celiac mac-rophage in vitro, degree of the phagocytosis enhanced with the drug concentration.Conclusion:1.The Sanzi Decoction have protective effect on mice infected with Staphylococcus or P.Aeruginosa. Significantly brought down the mortality rates of infected mice, the effective dose are 0.8g/kg, 0.4g/kg. Sanzi Decoction showed antibacterial effect in vivo, the research in order to provide theoretical proofs for the drug clinical application. 2. Compare with the control group, The three different dose (0.8g/kg, 0.4 g/kg, 0.2 g/kg) groups’thymus index and sp-leen index are not significant increase, the Sanzi Decoction can not effect the mice immune organ weight significantly. 3. In vivo, Sanzi Decoction was not showed significant efficacy of ConA induced spleen lymphocyte cell proliferation, but in vitro when it cultured with spleen lymphocyte cell, the drug can promote the proliferation of ConA induced spleen lymphocyte cell and normal spleen lymphocyte cell. in this experiment the optimal concentration are 25 ug/ml, 50 ug/ml, 100ug/ml. Suggesting that Sanzi Decoction showed mitogen function and synergistic with ConA in vitro. 4. In vivo, Sanzi Decoction can enhance the function of mice celiac macrophage phagocyte red blood cell. Cultured the mice celiac macrophage with the drug in vitro, it also can promote the phagocytic function, Sanzi Decoction can enh-ance the animal non-specificity immune function.
