Abstract


Objective:To investigate the effects of synergistic anti-gastric cancer effect of cytokine Induced killer cells pulsed with gastric cancer antigen loaded Dendritic cells in combination with Newcasatle Diease virus.Methods:Mononuclear cells were isolated from peripheral blood of healthy adult to induce CIK cells and DC cells.The phenotype of CIK cells and DC cells were detected by flow cytometer.Human gastric cancer cells SGC-7901 were cultured in vitro and the antigen suspension of cancer cells were prepared by repetitive freeze thaw method.Then the antigen was used to allergize DC cells.CIK cells were cultured in vitro with allergized DC cells.5×10~7 gastric cancer cells were injected subcutaneouly into right inguinal groove of hairless mice to establish the animal model,then the mice were separated into 4 groups.①PBS was injected into tumors;②NDV were injected into tumors;③Ag-DC-CIK were injected into tumors;④Ag-DC-CIK were injected into tumors after NDV.The volume of tumor was measured to calculate the tumor inhibitory rate.At the end of the experiment the tumors were resected and made into pathological section,then necrosis area scores were noted after HE stain.The experimental data were combined to make the factorial analysis.Results:1.NDV group:The average volume of tumor was 2363.298 mm~3 and the tumor inhibitory rate was 19.69%.The score of tumor necrosis area was 0.833.2.Ag-DC-CIK group:The average volume of tumor was 2230.405 mm~3 and the tumor inhibitory rate was 24.21%.The score of tumor necrosis area was 1.167.3.NDV +Ag-DC-CIK group: When gastric cancer cells were modified by the NDV first,the tumor inhibitory rate of Ag-DC-CIK increased to 67.29%.And the average volume of tumor was 982.330 mm~3. The score of tumor necrosis area was 2.833.We found that NDV and Ag-DC-CIK has a synergistic effect to kill gastric cancer cells(P＜0.05).Conclusion:1.NDV or CIK pulsed with gastric cancer antigen loaded DC has a weak effect to kill gastric cancer cells.2.CIK pulsed with gastric cancer antigen loaded DC combination with NDV have a synergistic effect to kill gastric cancer cells,obtained the best effect.
