Abstract


Objective: To observe the effects of the proliferative inhibition of human CNE-2Z cells on nano-TiO2 under photocatalysis,in order to investigate the mechanism of killing human CNE-2Z cells by nano-TiO2 under photocatalysis.Methods: The inhibition of proliferation of CNE-2Z cells ,which were treated by different concentrations of DMSO,and treated by different concentrations of nano-TiO2 and different photo- irradiation times,were analysed by MTT assay in vitro. The cells apoptosis was observed by transmission electron microscope（TEM）. Bax mRNA expressions of CNE-2Z cells were detected by reverse transcriptase-polymerase chain reaction（RT-PCR）,which were both before and after treated by different concentrations of nano-TiO2.Results:①The effect of proliferation of CNE-2Z cells was not significant when the concentrations of DMSO was between 0.05%～0.2%,but was evident between 0.5%～2%.②The Nano-TiO2 treatment obviously inhibited the proli- feration of CNE-2Z cells under photocatalysis in vitro,and the inhibition of proliferation is more obvious with the nano-TiO2 concentration and photo- irradiation time increases.③We observed that the nano-TiO2 could accelerate apoptosis of CNE-2Z cells under TEM. besides,we could observe the nano-TiO2 particles enter into inner cells.④The expression of Bax mRNA were weakly positive in CNE-2Z cells before nano-TiO2,but significantly enhanced in higher concentrations after nano-TiO2 under photoc- atalysis.Conclution: Nano-TiO2 can inhibit the proliferation of CNE-2Z cells under photocatalysis,which shows concentration and photo- irradiation time dependence. The induction of apoptosis maybe one of its mechanism.The results provided reliable experimental evidence for applying nano-TiO2 to clinical treatment of nasopharyngeal carcinoma.
