Abstract


Objective: Cervical cancer is the most common gynecologic malignancy, a serious threat to women's health and lives. In recent years, a steady increase in the incidence of the crowd and getting younger and younger. How early diagnosis and treatment of cervical cancer is important and we face a daunting task. Cervical cancer have a more lengthy precancerous stage, which for its early diagnosis and treatment of the conditions. Currently investigate the incidence of cervical cancer, the development of mechanisms has become a research hotspot. Cervical cancer, development is a multi-factor, multi-stage result of the interaction, the activation of cellular oncogenes and tumor suppressor gene inactivation is an important mechanism for cervical cancer. PTEN (phosphatase and tension homologue deleted on chromosome ten) is by far the first one found with phosphatase activity of tumor suppressor genes. PTEN gene in tumor incidence, progression, metastasis and prognosis of research has become a hot issue. PTEN gene and cervical cancer is currently no definite conclusion. This study was detected by immunohistochemistry chronic cervicitis, cervical intraepithelial neoplasia (cervical intraepithelial neoplasia, CIN) and invasive cervical squamous cell carcinoma of PTEN expression in order to further clarify the incidence of cervical cancer, the development of mechanisms, Early cervical cancer diagnosis and treatment, relapse prevention, provide a theoretical basis for improving the prognosis. Methods: Third Hospital of Hebei Medical University, Department of Pathology, January 2000 - December 2006 saved 110 cases of cervical paraffin-embedded tissue, including 30 cases of chronic cervicitis, cervical intraepithelial neoplasia (cervical intraepithelial neoplasia CIN) Ⅰ , Ⅱ, Ⅲ class organizations were 25 cases, 15 cases, 10 cases of invasive cervical squamous cell carcinoma 30 cases (Ⅰ 10 cases, Ⅱ of 10 cases, Ⅲ ~ Ⅳ 10 cases), patients aged 25 to 73 years mean age 35.5 years, all patients were not receiving the specimens before radiotherapy and chemotherapy and immunotherapy, all tissue specimens by two pathologists diagnosis. Immunohistochemistry was used to detect PTEN in 110 cases of cervical lesions in different tissues. SPSS13.0 statistical software, OK χ2 test, Fisher's exact test, test level α = 0.05. Results: chronic cervicitis, CIN Ⅰ, CIN Ⅱ, CIN Ⅲ, invasive cervical squamous cell carcinoma PTEN expression rates were 100%, 92%, 73.33%, 40%, 36.67%. In the above-mentioned organizations, PTEN positive expression rate gradually decreased, and the difference was statistically significant (P lt; 0.05). CIN Ⅱ, CIN Ⅲ, respectively invasive cervical squamous cell carcinoma and chronic cervicitis group, the difference was statistically significant (P lt; 0.05); CIN Ⅲ, invasive cervical cancer group were compared with CIN Ⅰ group, the difference was statistically significant ( P lt; 0.01); CIN Ⅱ and invasive cervical squamous cell carcinoma group, the difference was statistically significant (P lt; 0.05); CIN Ⅲ group and invasive cervical squamous cell carcinoma group, the difference was not statistically significant (P gt; 0.05) . PTEN expression and pathological staging of cervical squamous cell carcinoma (P lt; 0.05). Conclusion: a chronic cervicitis, CIN Ⅰ, CIN Ⅱ, CIN Ⅲ, invasive cervical squamous cell carcinoma of PTEN expression decreased gradually, the difference was statistically significant (P lt; 0.05), indicates that PTEN and cervical squamous cell carcinoma, the development of relevant . 2 CIN Ⅱ, CIN Ⅲ, respectively invasive cervical squamous cell carcinoma and chronic cervicitis group, the difference was statistically significant (P lt; 0.05); CIN Ⅲ, invasive cervical cancer group were compared with CIN Ⅰ group, the difference was statistically significant (P lt; 0.01); CIN Ⅲ group and invasive cervical squamous cell carcinoma group, the difference was not statistically significant (P gt; 0.05), shows that early in the process of cervical carcinogenesis PTEN that is changed by detecting the expression of PTEN to cervical cancer screening in high-risk populations for early diagnosis of cervical squamous help. 3 PTEN expression and clinical stage of cervical squamous cell carcinoma, squamous cell carcinoma of the cervix may be evaluated as prognostic markers. 4 PTEN gene therapy of cervical cancer are expected as a target gene for clinical application.
