Abstract


Objective: To investigate infertility in recent years, immunological factors are becoming a hot research topic for Reproductive Medicine, in a number of immune factors in anti-sperm antibodies (Atisperm Atibody, AsAb) and anti-cardiolipin antibodies (Aticardiolipin Atibody, ACA) more attracted attention. The male sperm itself, as a hidden antigen, sperm for women is an alien antigens, but under normal circumstances under physiological protection mechanism, and no anti-sperm antibodies, when physiological protective mechanisms are destroyed after causes the body to produce anti-sperm antibodies. Anticardiolipin antibody is an anti-phospholipid antibodies, under normal circumstances, the negatively charged phospholipids located in the heart of the inner layer of the cell membrane lipid bilayer is not recognized by the immune system, once exposed, cardiolipin antigens that stimulate the body to produce a autoimmune antibodies that anticardiolipin antibodies. In this study, by detecting infertile couples three subtypes anti-sperm antibodies in serum IgG, IgM, IgA, and anti-cardiolipin antibodies three subtypes IgG, IgM, IgA investigate the expression of anti-sperm antibodies and anti-cardiolipin antibodies in infertile infertility in rats. Methods: a research object: Select from May 2008 to October 2008 at the Third Hospital of Hebei Medical University, Department of Urology and the Bethune International Peace Hospital outpatient andrology clinic infertile couples 50 pairs as the experimental group (males aged 25 to 31 years average 26.70 ± 1.67, females aged 23 to 32 years, mean 26.22 ± 3.05) were first detected in serum anti-sperm antibodies and anti-cardiolipin antibodies. Inclusion criteria: married a year or more, couples sex life is normal, did not use any contraceptive measures without a history of breeding couple. The man without a family history of genetic disease, no chronic diseases, urological examination found no testis, epididymis and vas deferens abnormalities. Wife has no family history of genetic disease, no chronic diseases, normal follicles with normal ovulation cycle, gynecological examination vagina, uterus, fallopian tubes and ovaries were normal. Normal control group: physical health have normal reproductive history of childbearing couples 40 pairs of non-pregnant state (males aged 24 to 31 years, mean 26.35 ± 1.85; women aged 23 to 33 years, mean 26.63 ± 3.47), urological and gynecological examination no abnormalities. 2 Specimen collection: Take fasting blood 3ml, no anticoagulation, the serum was separated and stored at -20 ℃ refrigerator tested. All men were selected for 5-7 days of abstinence, taking semen semen analysis. 3 Test method: enzyme-linked immunosorbent assay (ELISA) detection of serum anti-sperm antibodies and anti-cardiolipin antibodies expression. Enzyme-linked immunosorbent assay (ELISA) detection kit was purchased from Shenzhen Security Group Biological Engineering Co., Ltd., according to kit instructions experimental operation and the results were determined. 4 Statistical analysis: The resulting anti-sperm antibodies and anti-cardiolipin antibodies represent data using count data, using Fisher's exact analysis. Semen analysis results to x ± s said, using a group t test analysis; All data on PC using SAS8.1 statistical package for statistical analysis. To P lt; 0.05 as statistically significant indicators. Result: an anti-sperm antibodies and anti-cardiolipin antibodies Results: The positive rate of anti-sperm antibodies males compared with control group difference was statistically significant (30% vs 5%, P lt; 0.01) See Fig1 and Table3. Experimental group of female antisperm antibody positive rate compared with the control group, the difference was statistically significant (34% vs 12.5%, P lt; 0.05) See Fig1 and Table5. Experimental group males anticardiolipin antibody positive rate compared with the control group showed no significant difference (6% vs 5%, P gt; 0.05) See Fig1 and Table4. Experimental group of women anticardiolipin antibody positive rate difference compared with the control group was statistically significant (32% vs 10%, P lt; 0.05) See Fig1 and Table6. 2 semen analysis results: The experimental group of male and anti-sperm antibodies anticardiolipin antibody negative patients compared with the control group, anti-sperm antibody-negative and negative anticardiolipin antibodies compared with semen volume, PH value, sperm density and b class sperm showed no significant difference (P gt; 0.05); experimental group sperm motility compared with the control group, a low level of sperm, straight lines and curves slow speed, with statistical significance (P lt; 0.01) See Table7. Conclusion: an anti-sperm antibodies and anti-cardiolipin antibodies is an important cause of infertility immune factors, anti-sperm antibodies can affect fertility in both men and women, and anti-cardiolipin antibodies major damage to women's reproductive function. Two anti-sperm antibodies can be reduced semen quality caused infertility.
