Abstract


【Purpose】 explore the neural stem cells (NSC), olfactory ensheathing cells (OEC), hematopoietic stem cells (HSC), Schwann cells (SC) alone transplantation and neural stem cells and olfactory ensheathing cells joint transplantation (NSC OEC), hematopoietic stem cells and Schwann joint cell transplantation (HSC SC) completely transected spinal cord injury in rat spinal cord VIII Ⅸ in lamellar neuronal survival, and provide theoretical and experimental evidence for cell transplantation in the treatment of spinal cord injury. 【Methods】 animals sham surgery group (SG group) spinal cord transection surgery group (CG group), neural stem cells (NSC), olfactory ensheathing cells (OEC), hematopoietic stem cells (HSC), Schwann cells (SC) alone transplant group, and co-transplantation of neural stem cells and olfactory ensheathing cells (NSC OEC), the joint transplantation of hematopoietic stem cells and Schwann cells (HSC SC) group. Were cultured in vitro NSC, OEC, HSC, SC, three-dimensional orientation of the NSC, OEC, HSC, SC, NSC OEC, HSC, SC were transplanted into the full cord injury in chronic phase (after 14 days) rat spinal cord cephalic, scar and tail of side. Paraffin sections of the spinal cord after perfusion fixation, line hematoxylin staining and immunohistochemical staining were counted the spinal cord VIII, IX lamellar number and size of neurons; observation host nerve regeneration. Using the SPSS 11.5 software package for statistical data processing. [Results] 1, cultured cells were treated with nestin, P75, CD34, and S100 immunohistochemical staining identified positive reaction. 2, the group of spinal cord the Ⅷ board layer number of neurons found in CG significantly reduced compared to the SG group, the the OEC group and the SC group than in the CG group was significantly increased the NSC OEC with the HSC SC joint transplantation group than those in the CG group was significantly increased, while the HSC SC group than in the HSC group more the OEC group than NSC OEC group were statistically significant (p <0.05). Comparison, the number of neurons Ⅸ in lamellar CG group was significantly reduced compared to the SG group NSC group, OEC Group with the SC group than in the CG group increased, the NSC OEC and the HSC SC joint transplantation group than in the CG group was significantly increased HSC SC group HSC groups were statistically significant (p <0.05). 3, each group of spinal cord ventral horn neurons in area comparing Ⅷ lamellar CG group than in the SG group significantly reduced (p <0.05), but the individual transplant group with the the CG group are not statistically significant. HSC SC joint transplantation group than the HSC group, SC group increased significantly (p <0.05), but the the NSC OEC joint transplantation group with the CG group, NSC group, OEC group were not statistically significant. Ⅸ board layer neurons area CG group than the SG group was significantly reduced (p <0.05), NSC, OEC, SC group increased significantly compared with the CG group was statistically significant (p <0.05), and NSC OEC joint transplantation and NSC groups, OEC group the the HSC SC joint transplantation group than in the HSC group, SC group were significantly increased (p <0.05). 4,5-HT nerve fiber regeneration, CG leader into scar 220 μm the NSC team leader into scar 2460μm the OEC Leader into scar 560μm, the HSC team leader into the scar 1820μm the SC team leader into the scar 1770μm, NSC OEC team leader into scar 890μm, HSC SC Leader into scar 3365μm. CGRP nerve fiber regeneration, CG head into the scar 600μm, the NSC team leader into the scar 940μm, the OEC team leader into scar 460 μm, the HSC team leader into the scar 680μm, the SC team leader into the scar 2090μm, NSC OEC group not long into the scar from scar headend 130μm, HSC SC Leader into the scar 3700μm. [Conclusion] cord injury led to a host spinal cord VIII Ⅸ in lamellar neuronal death, the remnants of the neuron cell body atrophy. 2, OEC alone transplant, SC alone transplant, the NSC OEC and the HSC SC joint transplantation group than in the CG group, the HSC SC joint transplantation than HSC alone transplantation significantly improved host VIII Ⅸ in lamellar neuronal survival quantity, while the NSC alone transplanted only improve host Ⅸ lamellar neuronal survival alone. 3, NSC, OEC, HSC, SC transplantation alone can not effectively prevent the the Ⅷ board layer neuron atrophy. HSC SC joint transplantation group, respectively, compared to the separate HSC or SC transplantation effectively prevents the Ⅷ lamellar motor neuron atrophy. 4 NSC, OEC, SC alone transplant, NSC OEC, HSC SC joint transplantation (respectively as compared to the respective single-cell transplantation) can effectively prevent Ⅸ board layer neuron atrophy. 5, NSC, OEC, HSC, SC alone or combined transplantation of cell transplantation is conducive to the 5-HT and CGRP axonal regeneration.
