Objective: osteosarcoma bone is the most common primary malignant tumors, 75% of patients aged 10-30 years old, is a serious malignancy adolescence it highly malignant and invasive, and early transfer of blood occurred. Adjuvant chemotherapy and neoadjuvant chemotherapy the application greatly improve the survival rate of the patients, but still nearly half of the patients died of metastasis. Tumor invasion and metastasis is the main biological characteristics of malignant tumors, is also the main cause of death in patients with malignant. So how effective prevention and treatment of metastatic, is essential to improve the prognosis of patients with osteosarcoma. In recent years, anti-invasion mechanisms of tumor invasion and metastasis and transfer of therapeutic areas conducted extensive research and found that the extracellular matrix (ECM) has played a key role in the process of tumor invasion and metastasis. The experimental results demonstrate the strength of tumor cell invasion and metastasis induced proteolytic degradation of the ECM and basement membrane is closely related to, and matrix metalloproteinases (MMPs) is one of the most important group of proteases in tumor invasion and metastasis plays an important role in recent years, the study of MMPs and their inhibitors are very popular. RECK gene is a gene discovered by Japanese scholars, has a unique function inhibit the secretion and activity of MMPs, RECK gene expression and tumor invasion, metastasis and angiogenesis are closely related, and can inhibit a variety of post-transcriptional level MMP expression, thereby inhibiting tumor invasion and metastasis. Currently, research on MMPs and RECK mostly concentrated in the epithelium-derived malignant expression in osteosarcoma and related research at home and abroad rarely reported. In this study, immunohistochemical staining, and of RECK expression of osteosarcoma occurs, development and infiltration of matrix metalloproteinase -2 (MMP-2), transfer relationship, and discussed its clinical significance. Methods: In this study, select the archive paraffin resting sarcoma specimens of 46 cases, 20 cases of osteochondroma specimens. Osteosarcoma is divided into 3 groups according to the pathological classification; divided into 2 groups based on the presence or absence of invasion and metastasis. Expression of RECK, MMP-2 detection by immunohistochemistry SP method, results were statistically analyzed using SPSS11.5 statistical package. Applications rank sum test (Kruskal-Wallis Test, Mann-Whitney U) expression of MMP-2 expression of RECK in osteosarcoma site, clinical stage, each subtype, invasion and metastasis grouping variable differences, chi-square test (Chi-Square Test) compare expression of MMP-2, expression of RECK in osteosarcoma, and MMP-2 expression of RECK single expression with invasion and metastasis of osteosarcoma, paired chi-square test (McNemar Test) method to measure the column contact number. P lt; 0.05 considered statistically significant. Results: MMP-2 in osteosarcoma positive rate was 65.22%, the positive rate was 25.00% in osteochondroma controlled organization, there are significant differences between both expression and MMP-2 in osteosarcoma Expression was significantly higher than the the osteochondroma control organization. Enneking staging of osteosarcoma continues to increase, MMP-2 positive and strong positive expression rates were gradually increased, there are significant differences in expression between the three different parts of the group in different gender groups, different pathological type group, MMP The -2 was no statistically significant difference. Significant MMP-2 expression in the presence or absence of invasion and metastasis, MMP-2 expression of invasion and metastasis group was significantly higher than the invasion and metastasis group. RECK in osteosarcoma positive rate was 41.30%, and 70% of the positive rate in the the osteochondroma control organization, differences between both expression was significant that RECK expression in osteosarcoma was significantly lower than osteochondroma control tissue. RECK positive and strong positive expression rate increasing with osteosarcoma Enneking staging descending order, there are significant differences in expression between the three different age groups, different parts of the group, different pathological type group, RECK expression no significant difference was that the expression of RECK strength and sex of the patient, tumor genotyping unrelated. In the presence or absence of invasion and metastasis, the expression of RECK significant RECK in expression of invasion and metastasis group was significantly lower than the invasion and metastasis group. In the same osteosarcoma samples, MMP-2 and RECK expression among the three groups were significantly different, higher RECK negative tendencies that MMP-2 positive. Single expression of MMP-2 and RECK determine whether the osteosarcoma invasion and metastasis among the three groups, there are significant differences in MMP-2 positive RECK negative higher invasion and metastasis tendency. Conclusion: MMP-2 in osteosarcoma generally high expression, RECK generally low expression; the osteochondroma RECK generally high expression, MMP-2 generally low expression. Pathological type and gender 2.MMP-2 and RECK expression with the osteosarcoma constitute diseased parts with soft tissue invasion and distant metastasis, clinical Enneking stage. 3.MMP-2 and RECK expression in osteosarcoma have some relevance. The balance between the 4.MMP-2 and RECK expression an important role in the process of invasion and metastasis of osteosarcoma, MMP-2-positive and RECK negative tendency has significant soft tissue invasion and distant metastasis. The joint detection of RECK and MMP-2 is expected to be one of the molecular targets for osteosarcoma of early diagnosis and prognosis.
