Abstract

Background and purpose of ischemic stroke as a clinical crisis, one of the urgent, serious illness, the incidence of both complex pathological basis, there are dangerous changing evolution, not only to the lives of patients poses a serious threat, and limb movement disorder, sequelae like language barriers, cognitive decline, but also to the patients themselves, family and society brought a heavy burden. In our country, as the socio-economic development, people's living standards continue to improve and the arrival of the aging society, the incidence of this disease is increasing year by year, so depth study of the pathogenesis of stroke, actively exploring effective treatment is a clinical workers and the common goal of scientists. Acupuncture treatment for several thousand years of history in our country has been related records as early as in the \function, swallowing function, is still widespread clinical application. Experimental studies confirmed that the experimental acupuncture can improve the neurological symptoms of cerebral ischemic injury in animal defects, reduce hypoxic-ischemic tissue damage and promote repair and functional reconstruction of the damaged area. Early resumption of the ischemic penumbra blood circulation, improve local hypoxia, is the focus of stroke treatment process to prevent further aggravation of the injury, the experiment as the starting point from ischemic injury, observe the change of the local the JAKS expression of combined The recovery of limb function, to verify the therapeutic effect of acupuncture, and to explore the possible mechanism of action. Modeling using the method of MCAO, to select the fuser closed brain artery induced rat model of focal cerebral ischemia. Observed after 2 hours, 24 hours, three days of each period the rat brain Jak1 level changes in apoptotic index and behavioral changes, as well as electro-acupuncture Baihui, Ojo above indicators, and to explore acupuncture regulation related mechanisms. . The reference Berderson and Belayev the neurological score after 2h, 1d, 3d paralyzed limb movements, in accordance with 0-4 points were assessed, the higher the score, the more severe injury. 2. Groups of nerve cells apoptosis index by TUNEL assay. Immunohistochemical detection of acupuncture pretreatment brain ischemia 2h, 1d, 3d damage zone brain the JAK1 level expression control study intervention effect. Results (1) neuromotor function scores 2h after the model group and acupuncture group that began to varying degrees of neurological defect signs, there is a significant difference (P <0.01) compared to the sham group, no significant difference between the two groups (P > 0.05); 1d, rats showed more closing side of the forelimb pronation or within, the damage to the contralateral side extrusion decreased muscle strength, some circling walking or occasionally circling walking; model group and EA group was significantly differences (P <0.05); 3d model rats symptoms of nerve defects reduced, there is a significant difference (P <0.01) with the sham group, EA group recovered significantly, with the sham group, there are still differences (P < 0.05); electro-acupuncture group and model group differences (P <0.05) (2) neuronal apoptosis index sham group at each time period showed no apoptotic cells; 2h, model group, EA group, see a small number of apoptotic cells were no significant difference (P> 0.05); 24h, model group and the electric needle group can be seen a large number of TUNEL staining positive cells, with the sham group comparison there is a significant difference (P <0.01), but EA group was significantly less in the model group (P <0.05); the 3d EA group TUNEL-positive cells decreased, electro-acupuncture compared with the model group, there is a significant difference (P <0.05). (3) JAK1 level detection sham group showed no expression, model group cerebral ischemic injury after 2h see the expression of a small amount of visible expression was significantly increased, 24h, expression continue until three days still, the EA group 2h see a small amount of expression, 24h seen significantly increased expression of the third day of the expression level higher than that in the model group (P <0.05) Conclusion ischemic brain damage caused by rats obvious signs of neurological deficit, nerve cells in the brain tissue apoptosis, electro-acupuncture stimulation Du \rat model has more obvious the JAK1 protein expression after experimental cerebral ischemia rat model after electro-acupuncture treatment the JAK1 protein expression increased consider electroacupuncture treatment of experimental cerebral ischemia rat model may by the JAK1 protein expression and phosphorylation of benign regulation play a role. One of the mechanisms may be cerebral ischemia induced JAK1 protein expression, electro-acupuncture group were significantly increased activity of JAK1 protein induced gene expression, and to reduce the occurrence of neuronal apoptosis and protective effect on cerebral ischemic injury .
