1. Clinical distribution and resistances to antibiotics of bacterial biofilmObjetiveIn order to understand the biofilm bacteria’s distribution, and their drug-ristance variability to floating bacteria, double-J tubes were selected for bacteria collection in our study. For further understanding of antibiotic-risistance of biofilm bacteria.Methods(1) Screening and identification of biofilm bacteria.92 urinary tract double-J tubes were collected from February to June 2009 in the Third Xiangya Hospital, Biofilm bacteria and their corresponding floating ones were screened and analyzed for their clinical distribution.(2) Comparison in variability of drug-resistance. Variability of drug-resistance of biofilm bacteria and floating ones were detetected on Mueller-Hinton culture medium. Variability of drug-resistance of biofilm bacteria were detetected on Poloxamer culture medium and MH culture medium as well.Results41 strains (45%) of biofilm bacteria were selected by Congo red test. Most of them were Enterococci(46.3%) and plasma-coagulase negative Staphylococcus(38.8%), the rest were Gram-negative bacteria(14.6%). There is no significant statistical difference of drug-resistance between biofilm bacteria and floating ones; The statistical difference of drug-resistance in biofilm bacteria on poloxamer and MH culture media is significant.ConclusionsEnterococci and Staphylococcus were the major biofilm bacteria, Poloxamer could simulate biofilm to reinforce its drug- resistance. 2. The correlation research of biofilm of Enterococcus faecalis and its related genes.ObjectiveIn order to detecte the formation of biofilm, gelatinase gene-gelE and pilus operon-ebpA, as Enterococcus faecalis related genes, were studyed.MethodsRT-PCR and real-time PCR were used to detecte gelE and ebpA genes in biofilm bacteria.ResultsEnterococcus faecalis, as a major pathogenic bacteria in hospital, was closely related to biofilm. The expression of ebpA of biofilm is 2019 times of floating ones, and that of gelE is 1/138 times.ConclusionsebpA is a positive factor for the biofilm fromation, and gelE is a negative one to restrain its formation.
