Abstract


Objective: To compare root canal re- treatment of root canal patients Enterococcus faecalis virulence factors gelatinase (gelE) expression analysis of Enterococcus faecalis virulence factor gelE and clinical performance relationship . Methods: 151 of the 131 patients need root canal treatment teeth ( each teeth only take a root canal ) , record the teeth of clinical symptoms , signs and information . Disinfection of teeth , sterile dental drill to remove the original filling , sterile root canal file to remove root canal filling . Reference to the X-ray image of the total length of the root canal from the apical 2-3mm lower end of the TDC , the sterile paper points enclosed the root canal 60s samples of the bacteria in the root canal . Extract samples of genomic DNA , polymerase chain reaction detection of Enterococcus faecalis . DNA enzyme treatment and extraction faecalis positive cases clinical samples of total RNA, total RNA by reverse transcription polymerase chain reaction to obtain cDNA . Real Time Quantitative PCR to detect of virulence factors gelE the expression . Statistical analysis of the relationship between the expression of virulence factors gelE clinical symptoms . Results: 1. Detection and identification by the polymerase chain reaction , 151 re- treatment of 58 cases of root canal cases were detected in Enterococcus faecalis detection rate of 38.4% . 2 Real Time Quantitative PCR detection of root canal re-treatment of 58 cases of E. faecalis of virulence factors gelE expression detected 57 cases the detection faecalis virulence factors gelE , expressed protein was 98% . Which enhanced expression of the 49 cases , decreased expression of the eight cases , patients without detection . The statistical analysis revealed treatment The root canal Enterococcus faecalis virulence factors gelE expression cases enhanced clinical symptoms or signs and apical Shadow closely related (P lt; 0.05), in cases of clinical symptoms or signs and the apical Shadow case gelE expression enhanced (P lt; 0.05). Conclusions: Enterococcus faecalis virulence factors gelE expression and clinical manifestations are closely related, the enhanced expression cases with clinical symptoms or signs Enterococcus faecalis virulence factors gelE . 2 . , Virulence factors gelE , Enterococcus faecalis expression with the treatment of root canal periapical destruction in close contact , enhanced expression of Enterococcus faecalis virulence factors gelE cases periapical destruction .
