Influences of Cisplatin on Expressions of P-glycoprotein in the Resistance Cells of in Lung Cancer Xenografts in the Mice

Abstract


Objective: To establish Lewis lung cancer drug -resistant cell line model, the mice were observed for the growth of xenografts and murine subcutaneous tumor cells P-glycoprotein expression , preliminary study of P- cell lung cancer in mice glycoprotein expression and cisplatin resistance relationship . Methods: C57BL / 6 mice were used as experimental animals , cultivate Lewis lung carcinoma cell line resistant to collect the logarithmic growth phase of the tumor cells , the concentration of the cell suspension adjusted to 1 × 107/ml. Mice were injected subcutaneously in the left hind leg cell suspension 0.2ml ( total number of cells was 2 × 105/ml), the tumor grew to 108mm when to start the experiment . The 40 mice were randomly selected from 10 , using immunohistochemical assay in mice transplanted tumor cells the expression of P-glycoprotein . The remaining 30 mice were randomized into computer cisplatin and saline groups , each group 15 . Cisplatin intraperitoneal injection of cisplatin 0.2ml ( administered in accordance with 1ml/kg ) / only the next day, a second , a total of four times ; saline group received saline 0.2ml / only intraperitoneal injection every other day for a total of four times , every two days after treatment length measuring mouse xenograft diameter , depicting tumor growth curve . 10 days after treatment, all the mice were sacrificed and immunohistochemical assay saline and cisplatin in mice transplanted tumor cells the expression of P-glycoprotein . Results: The tumor formation in mice was 100% before treatment mouse P-glycoprotein positive rate was 20% ( 2 /10 ) , saline group of P-glycoprotein -positive rate was 20% ( 3/15 ) cisplatin group P-glycoprotein positive rate was 33.3% ( 5 / 15 ) . Cisplatin group, the average inhibition rate was 31.29% . Cisplatin on tumor growth in mice significantly inhibited , compared with the saline group had significant difference (P lt; 0.05). Cisplatin group of P-glycoprotein expression was significantly higher than the saline group, the difference was statistically significant (P lt; 0.05). Conclusions : 1. Cisplatin in mice transplanted tumor cells showed a higher P-glycoprotein positive expression , suggesting that cisplatin treatment resulted in many tumor cells and drug- resistant produce aggravation : 2.P- sugar protein expression levels predict lung cancer resistant to cisplatin one marker , thereby providing individualized treatment of lung cancer reference index .
