Abstract


Ran GTPase protein belongs to a class of small G protein superfamily members, mainly distributed in the nucleus, these proteins has been shown to be involved in the material transport system between the nucleus and cytoplasm. However, Ran GTPase-protein's role in the immune system has remained unclear. Our study found that the amount of transcription of shrimp Ran GTPase (named PjRan) in disease-resistant shrimp and expression levels relative to the expression of normal shrimp (non-disease-resistant shrimp) was significantly higher, suggesting PjRan protein may be involved to shrimp against viral infections The innate immune system. Based on the study of the interaction between the protein, we found that the PjRan protein myosin protein interactions and the formation of protein complexes by actin myosin protein involved in cell phagocytosis process, so PjRan-myosin protein complexes may be swallowed related. In order to understand the role of PjRan protein in shrimp phagocytosis, on the one hand, using RNA interference technology knock low PjRan gene expression to detect changes in the prawns blood phagocytic; other hand using PjRan overexpression analysis, to study the impact of phagocytosis. The results showed that, after inhibit PjRan gene expression by RNA interference technology, the shrimp blood cell phagocytosis decreased significantly; contrast, the increase in the amount of PjRan gene expression shrimp blood phagocytic activity was significantly enhanced. The study shows that, PjRan gene silencing led to a significant increase in copy number of Shrimp White Spot Syndrome Virus (WSSV) PjRan gene overexpression resulted in a significant decrease in the copy number of WSSV. The above results show that PjRan protein through the regulation of the shrimp blood cell phagocytosis process to participate in the shrimp antiviral immune.
