Studies on Polymorphisms of LEPR and ZAG Genes and Their Associations with Growth Traits in Cattle

Abstract


In this study, using PCR-SSCP and DNA sequencing technology to detect the LEPR gene Nanyang cattle, cattle, Jiaxian Red, Jinnan cattle, Angus and Holstein six groups of 655 individuals and ZAG gene genetic variation analysis of its genetic structure and genetic diversity, and Nanyang cattle, Qinchuan and jiaxian, Red Bull three groups in the loci polymorphism and growth traits associated analysis to test these genes polymorphism cattle growth and development of genetic effects, in order to found a significant effect of genetic markers for economically important traits, genetics basis to establish a database for efficient breeding cattle and molecular markers, germplasm conservation and utilization. We obtained the following results: 1.6 cattle breeds in LEPR gene polymorphism were detected in 6 LEPR gene locus L1 ~ L6, containing exon 2 to exon 7, 6 genes in LEPR gene seat, only polymorphism was detected in the L3 loci L1 and L2, L4 to L6 loci found no polymorphism. L3 locus for the first time in five varieties of Nanyang cattle, cattle, Jiaxian Red Jinnan cattle, Angus found five SNP (NC 0 07301: g.26767T gt; C, 26805C gt; T, 27050A gt; G, 27063G gt; A and 27079G gt; A), 5 SNPs complete linkage, two SNP in intron 3, after three SNP located outside exon 4 on which gt 27050A; G and 27079G gt; to A mutation respectively lead LEPR protein 197His gt; the Arg and 207Asp gt; Asn amino acid mutations. Found no polymorphism in Chinese Holstein. The chi-square test Jiaxian Red in Hardy-Weinberg non-equilibrium state, while the remaining five species in equilibrium. Nanyang cattle, cattle, Jiaxian Red, Jinnan cattle Angus cattle and Chinese Holstein population genetic polymorphic index Ne / PIC / He 1.56/0.29/0.36 and 1.83/0.35/0.45 1.82/0.35/0.45 and 1.68/0.32/0.41, 1.82/0.35/0.45, 1.00/0.00/0.00. The genotype distribution of the chi-square test for independence, the Chinese Holstein with local cattle and Angus cattle difference highly significant (P lt; 0.01). The associated the 2.LEPR gene polymorphism and growth traits of Nanyang cattle, Qinchuan and jiaxian, Red Bull L3 locus analysis in cattle, LEPR gene found three genotypes of AA, AB, BB. In the in Nanyang cattle population, the AA genotype 6 month weight, average daily gain was significantly greater than the BB genotype (P lt; 0.05); AA genotype 6-month-old body height, body length was significantly greater than AB and BB individuals (P lt; 0.05 or P lt; 0.01), AA-and AB genotype 6-month-old bust was significantly greater than the BB genotype (P lt; 0.05). AA genotype 12-month-old body height, body length was significantly greater than AB and BB genotype (P lt; 0.05 or P lt; 0.01); AA-and AB genotype was significantly greater than the BB genotype (P 12-month-old bust lt; 0.05 or P lt; 0.01). Jiaxian Red groups, the AA genotype Bust, girth is significantly greater than the AB genotype (P lt; 0.05). Found the Qinchuan in no locus polymorphism was significantly associated traits. 3.6 cattle breeds in the ZAG gene polymorphism analysis of ZAG gene were detected in the Z1, Z2, Z3 and Z4 four loci, except Z2 locus, the other three loci found a polymorphism. First detected in cattle the six SNP (NC 0 07326: g 115T gt; C, 3257G gt; A, 4013G gt; A, 4027C gt; T, 4032T gt; C and 4120C gt; T), all located in the coding region of which 115C gt; T mutation leads to Ser 17 mutated to Pro 3257A gt; G mutations lead to 189 Arg mutation of Lys 4013A gt; G mutations lead to 233 Gly The mutation of Glu, 4027T gt; to C mutations lead to 238 Arg mutation Trp 4120T gt; C mutations lead to the 269 Pro mutation of Ser. Z1 loci found only in Chinese Holstein polymorphism. Nanyang cattle, cattle, Jiaxian Red, Jinnan cattle Angus cattle and Chinese Holstein six groups Z1 locus genetic polymorphic index Ne / PIC / He 1.41/0.25/0.29, 1.48/0.27 / 0.32,1.47 / 0.28/0.32 1.34/0.22/0.26 1.48/0.27/0.32 1.36/0.23/0.27; Z3 polymorphic loci index Ne / PIC / He 1.25/0.18/0.20, 1.06/0.06 / 0.06,1.05 / 0.04/0.05 1.03/0.04/0.03 1.14/0.11/0.12 1.00/0.00/0.00; the Z4 locus genetic polymorphism index Ne / PIC / He 1.52/0.30/0.34, 1.47/0.28 / 0.32,1.48 / 0.29/0.32 1.39/0.27/0.29 and 1.27/0.17/0.18 1.00/0.00/0.00. Z1 and Z4 locus in most groups, a moderate polymorphism Z3 locus of all groups in the low degree of polymorphism. The genotype distribution of the chi-square test for independence in Z3 and Z4 loci of Chinese Holstein with local cattle and Angus cattle difference was significant (P lt; 0.05 or P lt; 0.01) 4.ZAG gene polymorphism associated with growth traits of Nanyang cattle, Qinchuan and jiaxian, Red Bull analysis in the ZAG gene Z1 and Z3 loci were detected for the two genotypes of AA and AB, AA, AB, AC three genes in the the Z4 locus detected type. By least-squares analysis, Z1 and Z3 polymorphic loci growth and development traits of Nanyang cattle, Jiaxian Red Qinchuan significant impact ZAG gene polymorphic loci Z4 the Nanyang cattle 18 months of age ischial end Wide have a significant effect on the AC-type individuals significantly greater than the Type AB individuals (P lt; 0.05); Jiaxian Red Bull AC type individual body oblique length, bust and body weight significantly with greater than AA, AB-type individuals (P lt; 0.05 or P lt ; 0.01); growth and development traits the Z4 loci Qinchuan no significant impact (P gt; 0.05).
