Objective: To explore the association between the occurrence of hepatocellularcarcinoma and the genetic polymorphism in HLA-DRB1, HLA-DQB1.Methods: Thirty-two hepatocellular carcinoma patients diagnosed throughpathology in Dalian area from2011to2012were collected as experimental group,22cases were male and10cases were female, aging from52to82. DNA was extractedfrom intravenous blood samples which were anticoagulated with EDTA. Andpolymerase chain reaction sequence specific primer(PCR-SSP) was used to typeHLA-DRB1and HLA-DQB1. The HLA test control group were consisted of100unrelated healthy people(50men and50women) aging from20to45in Dalian area,andthe test data were provided by Dalian Friendship Hospital.That whether there weresignificant differences between the two groups in frequency of gene and homozygotewere analyzed by statistical methods.Results: HLA test results showed that: the experimental group of HLA-DRB1,HLA-DQB1gene frequency detection showed that: the gene frequency ofHLA-DQB1*03was significantly higher than that of the control group, while the genefrequency of HLA-DQB1*06was significantly lower than that of the control group, andthere were significant differences in thattwo locus..While the gene frequency of HLA-DRB1*13is significantly lower than thatof the control group,there were no significant differences on the rests of gene locusbetween experimental group and the control group.Conclusion:The expression of HLA-DQB1*03and HLA-DQB1*06gene mightassociate with the genetic susceptibility to the risk of the development of hepatocellularcarcinoma. The lack or decrease of HLA-DRB1*13gene expression makehepatocellular carcinoma escape from the immune system more easily. So thatHLA-DRB1*13may be one of the hepatocellular carcinoma’s protector genes. HLA-DQB1*05/05has a much higher actual frequency than theoretical frequency, inwhich significant statistical difference (P<0.005) does exist. ThereforeHLA-DQB1*05/05may be one of the predisposing genes of hepatocellular carcinoma,increase the risk of the development of hepatocellular carcinoma.
