Analysis identified specific nuclear matrix protein associated with apoptosis of esophageal cancer, and to explore the role they are in the process of apoptosis in esophageal cancer, to further understanding of esophageal cancer cell apoptosis and its mechanism from a more holistic level, thus Find the new direction of the esophageal cancer cell apoptosis research. The experimental results show that after a 8μg/mL curcumin treatment, EC9706 cell proliferation was significantly inhibited cell growth inhibition rate was 69.9%; cell cycle hypodiploid (sub-G 1 of) cells peak, apoptosis rate of 23.0%, and G 2 / M phase arrest occurs; light microscopy and transmission electron microscopy results show EC9706 cells induced by curcumin treated cell shrinkage nucleocytoplasmic ratio decreases, nuclear condensation, nuclear chromatin condensation, swelling of mitochondria, endoplasmic reticulum lumen to expand, the formation of apoptotic bodies and other significant features of apoptosis; apoptosis-related proto-oncogene bcl -2 downregulation promote apoptotic gene p53, fas, bax upregulation. The selective extraction ready to light and electron microscopy showed that the curcumin treatment EC9706 cells, the nuclear matrix - intermediate filament system can maintain a more complete network structure, but the number and density of the fibers than the control group sparse and more unevenly distributed, and the presence of a local fiber breakage, nuclear matrix - intermediate filaments overall system configuration tends to disintegration; two-dimensional gel electrophoresis analysis showed 31 differentially expressed EC9706 before and after apoptosis induced by curcumin nuclear matrix proteins by mass spectrometry analysis identified 12 proteins, which in apoptosis of EC9706 cells express the upward or emerging protein 8: Ran-binding protein 3-like Mitochondrial 28S ribosomal protein S30, Rho guanine nucleotide exchange factor 19, Rap1 GTPase-activating protein 2, Hsp70-binding protein 1,2-5A-dependent ribonuclease, Adseverin, TATA elementmodulatory factor; downregulated protein 4: Probable peptidyl-tRNA hydrolase, ATP synthase mitochondrial F1 complex assembly factor 1, Serine protease hepsin, Cell division protein kinase 6. Among Adseverin outside, other proteins were first found in the nuclear matrix. The study results show that the 8μg/mL curcumin on human esophageal carcinoma EC9706 cell apoptosis has significantly induced in EC9706 cell apoptosis process, not only the nuclear matrix - intermediate filament system configuration obvious changes in the characteristics of apoptosis , and the composition of the nuclear matrix protein change significantly. Which confirmed the presence of specific nuclear matrix protein associated with esophageal cancer cell apoptosis and its role induced apoptosis of esophageal cancer.
