Abstract


Objective: To investigate the immunomodulatory effects of wood hoof Phellinus polysaccharide (Fomes fomentarius polysaccharides, FFP, wooden hoof polysaccharide) and its possible mechanism of action. Methods: MTT was used to detect the metabolic activity of the immune cells of normal mice, mouse peritoneal exudate light microscope drug effects changes in cell morphology and density, the ELISA assay tumor necrosis factor α (TNF-α), interferon-γ ( IFN-γ), interleukin-2 (IL-2) content, chicken erythrocyte phagocytosis assay macrophage phagocytic function, sheep red blood immunoassay detection of humoral immune function of normal mice. Subcutaneous injection of cyclophosphamide (cyclophosphamide, CP) copy immunocompromised animal models, weighing thymus, spleen weight viscera index carbon clearance method for the determination of the monocyte-macrophage phagocytosis, sheep red blood immunoassay immunosuppressive small mouse humoral immune function, while dinitrofluorobenzene (dinitro-fluorobenzene, DNFB) induced delayed type hypersensitivity (delayed-type hypersensitivity, DTH) model, observe the immune suppression function of immune cells, microscope counting method counts the outer periphery of mice blood leukocytes. Results: FFP (100,50,25 μg / ml) can significantly promote the metabolic activity of the spleen cells of normal mice (P = 0.000, P = 0.001, P = 0.004), to promote normal mouse spleen cells secrete cytokines TNF-α ( P = 0.000, P = 0.000, P = 0.001), IFN-γ (P = 0.000, P = 0.000, P = 0.001), IL-2 (P = 0.002, P = 0.002, P = 0.023), and the control group compared, FFP to promote the role of the spleen cells to secrete TNF-α is the most obvious; FFP significantly enhance the normal mouse peritoneal exudate cells metabolic activity (P = 0.000, P = 0.000, P = 0.004), promote peritoneal exudate cells secrete TNF- α (P = 0.000, P = 0.000, P = 0.000), and a dose-dependent trend in a certain concentration range. Light microscopy showed FFP administered group peritoneal exudate cells in cell density increased, the morphological change significantly, cells oval-shaped rod, fusiform, polygonal or irregular in shape, surface irregularities or pseudopodia-like protrusions. FFP (100,50,25 mg / kg) significantly enhanced sheep red blood cells (SRBC) generated due to normal mice specific antibodies (P = 0.005, P = 0.017, P = 0.044), FFP (100,50 mg / kg) can The enhanced phagocytic rate of normal mouse peritoneal macrophage phagocytosis of chicken erythrocytes (CRBC) and phagocytic index (P = 0.000, P = 0.002) (P = 0.000, P = 0.004). FFP high, medium dose (200,100 mg / kg) can significantly improve the cyclophosphamide-induced immune suppression in mouse thymus and spleen index (P = 0.003, P = 0.044) (P = 0.010, P = 0.038). FFP high dose (200 mg / kg) can significantly improve the cyclophosphamide induced immunosuppressed mice carbon clearance ability (P = 0.015), and improve immune function and inhibition of mouse macrophage phagocytosis. FFP high, medium dose (200,100 mg / kg) can significantly improve sheep red blood cells (SRBC) caused by cyclophosphamide-induced immunosuppressed mice serum hemolysin level (P = 0.005, P = 0.031). DTH reaction FFP high, medium dose (200,100 mg / kg) can significantly improve the cyclophosphamide-induced immunosuppressed mice ear swelling (P = 0.003, P = 0.029), the FFP of high dose (200 mg / kg) significantly improve cyclophosphamide-induced immunosuppression DTH reaction in mice ear swelling rate (P = 0.017). FFP high, medium dose (200,100 mg / kg), cyclophosphamide treated mice could increase the level of white blood cell count (P = 0.001, P = 0.033). Conclusion: 1. The the wood hoof polysaccharide (FFP) of normal mice with immune function, can significantly enhance the metabolic activity of normal immune cells, promote normal immune cells secrete cytokines TNF-α, IFN-γ, IL-2 and enhancement of the humoral immune function of normal mice. 2.FFP vitro IL-2, IFN-γ secretion, promote the overall level of normal mice produce hemolysin that IgM, suggesting enhanced humoral immune function mechanism may be related to the promotion of IL-2, IFN-γ secretion. The 3.FFP vitro can significantly enhance the metabolic activity of normal mouse peritoneal exudate cells (macrophages), can promote normal mouse peritoneal exudate cells secrete cytokines TNF-α, The FFP in vivo monocyte-macrophage activation and enhanced phagocytic function of the cells, suggesting that the macrophage is the the FFP role of one of the target cells. 4 cyclophosphamide successfully established in immunocompromised mice experimental model. 5. Wooden hoof polysaccharide on cyclophosphamide-induced immune suppression mice with immune function, a significant increase in the cyclophosphamide-induced immune suppression in mice immune organ index, increased its correction carbon clearance index (α) increase phagocytic activity, promote serum hemolysin antibody enhanced delayed-type hypersensitivity, increase the white blood cell count, and prompts the FFP can significantly promote cyclophosphamide induced immunosuppressed mice non-specific and specific (humoral and cellular immunity) immune function. 6 preliminary proof the wooden shoe polysaccharide is a good immunomodulatory agents.
