Abstract

The bees are social insects, induced nutritional factors, the female bee from larval stage to start gradually differentiate into two sibling type of the queen bee and the worker bees, bee populations exposed to different social functions. Female bee level differentiation mechanism is not fully understood, but studies have shown that the female bee level type differentiation in vivo DNA methylation level related. Nutrition and space factors in order to study the relationship between the female bee developmental impact of DNA methylation, the experimental study mellifera measured under different experimental conditions 3d, 5d and 6d larvae the head Dnmt3 enzyme activity, Dnmt3 mRNA The relative expression level, dynactin p62 gene methylation level, and just left the room individual birth weight, body length, forewing length, forewing width, snout length, third backplane long morphological index. The results show that: in the the consistent spatial factors, as the larvae feed royal jelly time reduction, the larval head Dnmt3 enzyme activity, Dnmt3 mRNA relative expression level of dynactin p62 gene methylation levels increased significantly, newly emerged female bees individual forewing length, forewing width, snout length significantly improved significantly, and birth weight, body length, the third backplane as long as a significant reduction. The decline in the proportion of larval development of queen bee larvae development toward the worker bees direction. Under the same conditions of nutritional factors, with the development of space decreases, the larvae head Dnmt3 enzyme activity and Dnmt3 mRNA relative expression level, dynactin p62 gene methylation level significantly increased, just feathering the female forewing length, forewing width snout length significantly increased, birth weight, body length, the third backplane as long as a significant reduction. The larval development worker bees increase in the proportion of larval development toward the worker bees direction. Injection Dnmt3 dsRNA through to 3d larvae head can significantly reduce the the larval head Dnmt3 activity of, Dnmt3 mRNA relative expression levels and dynactin p62 gene methylation level significantly improve the newly emerged female individual birth weight, body length, the third back plate length. The decline in the proportion of larval development worker bees, larvae and development toward the queen bee direction. These results suggest that bee workers and worker bees differentiation, nutrition and space factors have an impact, but the impact of dynactinp62 gene methylation sites are different. For the first time found that spatial factors can be regulated by DNA methylation female bees development, and further confirmed by the female bee class differentiation Dnmt3 closely related.
