Cruciferous annual The cotton of fruit Mosla (Lachnoloma lehnannii) is located in northern Xinjiang Junggar Desert Ephemeral Plants. The study was to evaluate the the the cotton fruit camelina tissue culture plant regeneration system to establish the key factors to early spring ephemeral plants cotton fruit camelina as test material, comprehensive system to explore some of the major factors that affect cotton fruit camelina tissue culture, cotton fruit camelina tissue culture plant regeneration, germplasm conservation and provide important reference. The test results show that: (1) cotton fruit camelina seed disinfection appropriate steps: 1 ~ 2 min under sterile conditions running water seeds sterile water washed four times soaked with 70% ethanol for 1 min, and then treated with 0.1% HgCl2 disinfection 1.5min, and finally washed 4 times with sterile water. (2) light on seed germination promoted, and different parts of the seedlings have a different response to light. Seed germination on low temperature dependence, while seedling growth requires a certain amount of high temperature. Appropriate hormones promote seed germination, and different parts of the seedlings have a different reaction to the hormone. The cotton fruit camelina seed germination and seedling growth for training system: the inoculated seed disinfection hormone-free MS at a temperature of 15/5 ℃ light conditions develop 4 to 5 d after germination Go to the temperature of 25 ± 1 ℃ cultured under the lighting conditions, the germination rate of 90%, 16% higher than the control, and the seedlings are growing well. (3) the cotton fruit Mosla callus induced appropriate sterile Miao Miao age 14d, no vaccine in different parts of the medium of a different hormone combinations on callus induction effect cotyledon callus induction effect good hypocotyl, cotyledon and hypocotyl highest callus induction rate in the treatment of 10mg / l IBA 0.1 mg / l BAP and 1.0mg / l IBA 0.5 mg / l BAP, callus induction rate of 100 % and 98%. Callus induction suitable medium for the high salt MS medium, suitable concentration of 3% sucrose. The callus induction without light, suitable temperature to 25 ℃ (4) cotton fruit camelina different sources callus growth suitable growth regulator with both MS 1.0 mg / l IBA 0.1 mg / lBAP but callus The different growing cotyledon and hypocotyl callus proliferation times of 5.3 and 2.5, respectively. The callus growth needs light, suitable temperature of 25 ° C. (5) the differentiation of the cotton fruit Mosla callus on growth regulating agent type, concentration and its complexes is particularly sensitive. Hypocotyl callus differentiation efficiency than the cotyledons. Hypocotyl and cotyledon callus differentiation optimum growth regulator with the MS 1.0 mg / lBAP 0.1 IAA and MS 2.0mg / l BAP 0.1 IBA differentiation rate of 91.7% and 80.9%, respectively, and the average number of adventitious buds 6.5 and 5.8. (6) cotton fruit camelina appropriate auxin rooting for the IBA, the best concentration of 1.0 mg / l, the processing rooting rate of 75.0%, average 4.6 root, root length of 3.0 cm. (7) cotton the fruit Mosla forging seedlings suitable for transplanting medium with perlite: vermiculite: sand = 1:1:2, the survival rate was 70.0%.
