Abstract

Objective of intestinal ischemia-reperfusion lung tissue and serum levels of tumor necrosis factor-α (TNF-α) and nuclear factor-κB (NF-κB) in the lung tissue impact, explore sufentanil sufentanil post-processing and possible mechanisms of rat intestinal ischemia-reperfusion lung tissue, to provide experimental basis for clinical. Methods 32 healthy adult male Wistar rats were randomly divided into four groups: sham operation group (S), intestinal ischemia-reperfusion group (I / R group), the treatment group after intestinal ischemia (I-post group) and 1ug/kg Sufentanil post-processing group (SP group). N = 8. ① sham group: only line open, separating the superior mesenteric artery (SMA) occlusion; ② ischemia - reperfusion group: occlusion of the superior mesenteric artery 60min prepared after perfusion 120min in intestinal I / R model; ③ ischemia treatment group: 60 min of ischemia immediately after perfusion line three cycles of 30 s / block 30 s, the same I / R group. ④ the sufentanil After treatment groups: of the 1ug/kg sufentanil diluted to 0.3ml reperfusion immediately into the femoral vein with a syringe pump pump 0.1ml/30S, during discontinuous 30s, so the cycle three times (a total of 0.3 ml), I with the I / R group. Reperfusion 120min killed eight rats. HE staining the pathological changes of the lung tissue of rats. The content of TNF-α in the serum and lung tissue were measured by radioimmunoassay; Determination of NF-κB in lung tissue level using immunohistochemistry. Results (1) HE detect pathological changes: ① S group: found no significant pathological changes and the alveolar size uniformity, the structural integrity of the thin wall, no bleeding and edema and inflammatory cell infiltration; ② the I / R group, uneven alveolar size, integrity The destruction of the alveolar space protein-like exudate, alveolar wall thickening interval widened pulmonary interstitial and alveolar edema, infiltration of inflammatory cells, hyperemia, hemorrhage significantly. ③ I-post groups, SP group above pathological changes were mild protein exudation reduce alveolar spaces, alveolar wall thickening, inflammatory cell infiltration mitigate (2) of TNF-α, NF-κB detection: compared with S group increase in the rest of the group of TNF-α, NF-κB upregulated (); compared with the I / R group, I-post group and SP group NF-κB downregulation, TNF-α reduced (P? 0.05); I-post compares the indicators of the group and SP group difference was not statistically significant (). Conclusions (1) intestinal ischemia and reperfusion can cause serious lung injury. (2) intestinal ischemia-reperfusion lung injury mechanisms is complex, its occurrence and development may be associated with TNF-α and NF-κB. (3) sufentanil treatment of intestinal ischemia-reperfusion lung injury caused by the protective effect, can inhibit NF-κB expression in the lung tissue, reduce serum and lung tissue of rat intestinal ischemia-reperfusion TNF- the α the content.
