Protective Effect of Edaravone in the Intestinal Mucosal Barrier Injury of Severe Acute Pancreatitis in Rats

Objective: To investigate the protective effect and mechanism of edaravone injury of rat intestinal mucosal barrier function in severe acute pancreatitis, which provide reference for the prevention and treatment of severe acute pancreatitis intestinal mucosal barrier dysfunction. Methods: 60 SD rats were randomly divided into normal group (Norm group) (n = 6), sham group (Sham group) (n = 18), severe acute pancreatitis (SAP group) (n = 18 ) and edaravone treatment group (ED group) (n = 18). Norm group without any treatment; abdomen was closed after the sham group only flip pancreas laparotomy; using SAP group SAP rat model of retrograde duodenal pancreatic duct injection of 5% sodium taurocholate solution preparation; ED group model were successfully instantly via the tail vein injection of the 3mg/kg the edaravone. After success in modeling, 6h, 12h, 24h blood, intestines, pancreas specimens. Detected in each group at different time points plasma diamine oxidase (DAO), serum amylase and intestinal tissue malondialdehyde (MDA), superoxide dismutase (SOD), intercellular adhesion molecule -1 (ICAM- 1) level, and light microscopy gut and pancreas pathological changes. Kolmogorow-Smirnow each group of data normality test fit the normal distribution with a mean ± standard deviation (x ± s) described single-factor analysis of variance (one-way ANOVA) line group differences between comparison If significant differences among the groups, further SNK test (q test) twenty-two comparison, correlation analysis using Pearson correlation analysis method. Results: 1.Norm group differences with Sham group, the plasma levels of DAO statistical significance (P gt; 0.05); SAP group rats after 6h, 12h, 24h plasma DAO concentration and serum amylase concentration were higher than Norm group and Sham group (P lt; 0.05); the ED plasma DAO concentration and blood amylase concentration compared Norm group and Sham group increased (P lt; 0.05), but lower compared to the SAP group (P lt; 0.05). Intestinal tissue 2.Norm group and Sham group MDA SOD level difference not statistically significant (P gt; 0.05); compared with the Norm group and Sham group, SAP group MDA levels three time points in turn increased (P lt; 0.05); 3 points of the activity of SOD in turn decreased (P lt; 0.05); ED group were significantly reduced compared with SAP group MDA water SOD activity was significantly higher (P lt; 0.05). Sham group, intestinal tissue ICAM-1 levels 3.Nrom group and the difference was not statistically significant (P gt; 0.05); compared with Nrom group and Sham group, SAP group intestinal ICAM-1 levels at three time points elevated (P lt; 0.05); the ED group intestinal ICAM-1 levels compared with SAP group decreased (P lt; 0.05), but still higher than Nrom group and Sham group (P lt; 0.05). 4.SAP group of intestinal histopathological changes include villi, lamina propria shedding, nudity, submucosal capillary congestion; pancreatic histopathological changes including severe edema, hemorrhage, necrosis, inflammatory cell infiltration; Norm group and Sham group pancreas intestinal score difference had no statistical significance (P gt; 0.05); the SAP group intestinal and pancreatic tissue injury score was significantly higher than Nrom group and Sham group (P lt; 0.05); the ED group intestinal tissue injury score than SAP There were no statistically significant (P gt; 0.05), but still higher than the Nrom group and Sham group (P lt; 0.05); the ED group pancreatic tissue injury score (P lt; 0.05) lower compared with SAP group, but still higher than Nrom group and Sham group (P lt; 0.05). 5 plasma DAO levels of intestinal MDA levels showed a significant positive correlation (P lt; 0.01), and intestinal SOD activity was significantly negatively correlated (P lt; 0.01); the plasma DAO intestinal tissue of ICAM-1 levels were significantly positively related (P lt; 0.01). Conclusion: SAP early that exist intestinal mucosal barrier dysfunction, edaravone has a protective effect on severe acute pancreatitis rat intestinal mucosal barrier dysfunction, the mechanism may be associated with downregulation of oxygen free radicals and cytokines levels and lowered endothelial cell ICAM-1 expression.
