Abstract


Objective: To study the α-tocopherol traumatic brain injury in the rat brain nerve growth inhibitor Nogo-A, NgR in protein expression, and to investigate the mechanism of action of nerve regeneration. : Weighing 250-300g healthy SD rats 120 randomly divided into sham group, traumatic brain injury group and α-tocopherol group, each group is divided into 1 day, 3 days, 7 days, 14 days and 21 days of five time points, the number of rats of each time point was 8. Moderate cerebral contusion in rats caused by traumatic brain injury group and α-tocopherol group were improved Feeney'S freefall model, sham group only open scalp and bone window, do not do to combat. α-tocopherol group administered six hours after injury, after daily intraperitoneal injection of α-tocopherol 200mg/kg, continuous injection 21d, traumatic brain injury group, the sham group injected with the appropriate dose of saline at the same time. Under mNSS score sheet of the three groups of rats at the corresponding time points after surgery neurological score. HE staining to observe pathological changes of the nerve cell morphology and immunohistochemical staining to detect changes in the nerve cells in Nogo-A and NgR protein expression, and comparative analysis. Results: 1. Histopathological changes: HE staining show a clear structure of the brain tissue of sham-operated rats, uniform and dense. Traumatic brain injury group, local hemorrhage, edema, and inflammatory cell infiltration, nucleus pyknosis dissolved, necrosis, after the gradual emergence of glial cells and fibrous tissue. Injection the а-tocopherol group is compared with brain trauma group have a certain degree of ease. 2. Compared with the sham group, traumatic brain injury rat brain Nogo-A protein expression at all time points were higher than the sham group (P lt; 0.01), 14 Telsda peak; compared with traumatic brain injury group Nogo-A protein expression in rat brain α-tocopherol group were lower at all time points (P lt; 0.05), statistically significant. 3. Compared with the sham group, traumatic brain injury rat brain NgR in the first day there reduced to a minimum on day 3 (P lt; 0.01), then gradually increased at 7 days with the sham group expression basically the same, 14 days, 21 days elevated compared with sham group (P lt; 0.01); treatment group NgR protein is basically the same trend with traumatic brain injury group and minimize (P lt; 0.05) in 3 days, 7 days basically the same with the sham group expression, in 14 days, 21 days, compared with brain trauma group was significantly reduced (P lt; 0.01), and statistically significant. Conclusion: 1. Α-tocopherol can effectively reduce the the Brain Injury organizations Nogo-A expression of NgR in promoting axonal regeneration. 2.α-tocopherol can significantly reduce traumatic brain injury in rats degree of injury, play a role in brain protection.
