ObjectiveTo investigate the changes of matrix metalloproteinase2and9 (MMP-2, MM- P-9) in rat cardiac hypertrophy,and the efects of Rosuvastatin(Ros) on it in a rat model.Methods40 Sprague-Ddwley SD rats were randomly divided into four groups including control group,Isoprenaline(ISO) group,Captoril(CAP) group and Ros- uvastatin(ROS) group,with 10 rats in each group.Cardiac hypertrophy models were induced by injection of ISO into rats of ISO group,CAP group and ROS group, Cardiac hypertrophy in rats was induced by subcutaneous administration of isoprenaline in which the dosage schedule was set to 20mg/kg in the 1st day,10mg/kg in the 2nd day,5mg/kg in the 3rd day , and 3mg/ kg in the following consecutive seven days while physiological saline were injected into control group,Then, ROS group were given ROS 4mg/kg intragastric administr- ation per day,CAP group were given CAP 60mg/kg intragastric administration per day,the other two groups were given physiological saline intragastric administration per day,After four weeks,Body weight(BW),heart weight(HW) and left ventricular weight (LVW) were measured,and their ratios(HW/BW,LVW/BW) were calculated, The contents of hydroxyproline(HYP) left ventricular were detected with spectrophotometry, then the contents of related collagen were calculated. The protein expression of MMP-2 and MMP-9 were examined by immunohist- ochemieal analysis.Results(1)The ISO group and control group set of comparison,bigger HW/BW and LVW/BW, the difference has statistics meaning (P<0.01), HW/BW of ROS group and CAP group is less than ISO group , the difference has statistics meaning (P<0.05); LVW/BW of ROS group and CAP group is less than ISO group (P < 0.01).(2) The ISO group and control group set of comparison,more HYP and the contents of related collagen,which are significance (P<0.01), HYP and the contents of related collage of ROS group and CAP group are less than ISO group(P<0.01).(3) Compared with the control group , ISO group showed much greater increase the protein expression of MMP-2 and MMP-9 (P<0.01); The protein expression of MM P-2 and MMP-9 of ROS group and CAP group were lower than ISO group prominent ly(P<0.01),(4) The efect of ROS group was similar to that of CAP group (P>0.05).ConclusionsROS can relieve the degree of myocardial fibering after cardiac hypertrophy, which was associated with the attenuated expression of myocardial MMP-2 and MMP-9.
