Abstract

Objective To study the Dan participate in the joint application of verapamil on hepatic ischemia-reperfusion injury in rats, to explore the two drugs in hepatic ischemia-reperfusion injury mechanism, Dan participation in the verapamil joint prevention and treatment of hepatic ischemia reperfusion injury in clinical applications provide experimental basis. Preparation of rat liver Nauta methods such ischemia-reperfusion model, were randomly divided into five groups: sham operation group (S), ischemia-reperfusion group (I / R group), Salvia preconditioning group (SM group), the verapamil pretreatment group (V group), Dan participation in the verapamil Joint preconditioning group (SV group), respectively, in hepatic ischemia reperfusion 2,8,24 h drawn, using immunohistochemical methods to detect liver organization of the nuclear factor-Kappa B (NF-κB), intercellular adhesion factor (ICAM) -1 expression, measured in the liver tissue myeloperoxidase (MPO) concentration evaluation of liver tissue neutrophil infiltration press kit method for measuring liver tissue malondialdehyde (MDA), superoxide dismutase (SOD) concentration, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and the level of light microscopy and electron microscopic examination of liver organization of the degree of cell damage. NF-κB A value in the I / R group reperfusion 2h (0.415 ± 0.042) was significantly higher, 8h (0.310 ± 0.031), 24h (0.244 ± 0.034) decreased. ICAM-1 A value in reperfusion 2h (0.359 ± 0.033) increased and reached the peak, 8h (0.450 ± 0.034), 24h (0.296 ± 0.022) decreased. Similar changes in MDA, MPO, ALT, AST and ICAM-l. SM, V, SV group indicators at each time point compared with the I / R group (P lt; 0.05), but lower than the S group (P lt; 0.05), and the SV group than in the SM, V group lower (P lt; 0.05). ALT, MDA, MPO, NF-κB and of ICAM-l between the changes have a positive correlation (P lt; 0.01). I / R group SOD in reperfusion reduce 2h (208.84 ± 16.84), 8h (184.80 ± 12.96) hit bottom, 24h (202.78 ± 14.70) elevated. SOD in the SM, V, SV group at each time point higher than in the I / R group (P lt; 0.05), but lower compared with the S group (P lt; 0.05), and the SV group than SM V group higher (P lt; 0.05). Changes between SOD and ALT, MDA was negatively correlated (P lt; 0.01). Light and electron microscopy no difference between the the SM group V group, among the rest of the group also has a significant difference. Conclusion hepatic ischemia reperfusion stimulate NF-κB activation, ICAM-1 expression and lower SOD activity participation HIRI occurrence; Dan to participate verapamil can significantly reduce ischemia-reperfusion liver tissue NF-κB activation, ICAM-1 expression and increased SOD activity effectively improve HIRI, and Dan synergies involved in verapamil combination of these effects.
