Abstract

Polymeric micelles is a novel pharmaceutical carrier , because of its unique structure and nature of the core-shell , in the field of nano - drug delivery system has a broad application prospect . The liver targeting amphiphilic chitosan derivatives - galactose palmitoyl chitosan preparation of polymeric micelles as carrier material , and study the physical and chemical properties and in vitro anticancer activity . Homemade our group with Varying Aliphatic degree of substitution of chitosan derivatives as a polymer micelle carrier material , emulsion - solvent evaporation method polymeric micelles were prepared and measured fluorescent probe polymer the micelles critical micelle concentration , screening the the appropriate CMC value galactose palmitoyl chitosan as the preparation of drug-loaded polymeric micelles material . Hydroxycamptothecin emulsion - solvent evaporation method , as a model drug , chloroform - ethanol mixed solvent to dissolve the drug and carrier material selected as the oil phase , the drug - carrier prepared polymeric micelles . Encapsulation efficiency and drug loading of the drug-loaded micelles was measured by UV spectrophotometry using orthogonal design experiments to examine the formulation and preparation process , on the basis of single factor experiments , the best optimized formulation and preparation process : drug / carrier ratio = 0.2:1 , the relative proportion of the oil / water = 1:7, the ultrasonic time = 40min . TEM observation drug-loaded micelles form, size detector determination of particle size and zeta potential . Release medium containing 0.02% Tween- 80 , 0.1 mol / L pH7.4 in PBS , visits loaded micelles in vitro release behavior , and analysis of the drug release mechanism . MTT assay, visits loaded micelles on human hepatoma cell line HepG2 growth inhibition in vitro , and compared with the hydroxycamptothecin solution , blank micelles , the results were lower than the drug-loaded micelles inhibitory effect on HepG2 cells hydroxycamptothecin solution , blank micelles have lower inhibitory effect on cell .
