Abstract


Objective: To observe the severe acute pancreatitis rat hepatic nuclear factor (NF-κB) activation and inducible nitric oxide synthase iNOSmRNA expression , and to investigate the protective effect of N-acetylcysteine ??(NAC) on liver injury of acute pancreatitis mechanism. Methods : 54 SD rats were randomly divided into sham operation group (SO ) , pancreatitis (SAP) and the NAC group (NAC group ) , 18 in each group . To 4% bovine bile sodium solution injected retrograde cholangiopancreatography production model of SAP . 1h femoral vein injection in the NAC group after modeling NAC (200mg/kg). 3h , 6h , 12h time point randomly killed rats . Using reverse transcription - polymerase chain reaction (RT-PCR) assay liver tissue iNOSmRNA expression the the SP immunohistochemistry assay NF-κB activity in liver tissue ; ; pancreas, liver biopsy tissues observed simultaneously serum amylase , liver function (AST, ALT) and MDA inspection. The : SAP group made ??mold NF-kB significantly elevated liver the organization iNOSmRNA expression and elevated serum malondialdehyde content , at each time point SAP group than in the SO group was significantly higher ( P lt ; 0.05 ) , the NAC group than SAP group was significantly lower ( P lt; 0.05 ) , the NAC group amylase , AST , ALT and MDA content significantly decreased compared with the SAP group ( P lt; 0.05 ) . Conclusion: The liver tissue NF-κB activation and iNOSmRNA overexpression may be one of the mechanisms of acute pancreatitis complicated by liver damage , NAC may inhibit hepatic NF-κB activity thereby inhibiting expression of iNOSmRNA , has a protective role in acute pancreatitis and liver injury .
