Abstract


Objective: To investigate the artificial synthetic phosphorothioate oligodeoxynucleotide (ODN) containing CpG motifs as adjuvant PBMC from chronic HBV infection, CD4 ~~ T lymphocytes, CD8 ~ T lymphocytes and DC and other immune cells affect the immune response, to further clarify the chronic HBV immune pathogenesis, and provide the experimental basis for new methods of immunotherapy for patients with chronic HBV hepatitis. Method: The outer periphery by density gradient centrifugation, from randomly selected chronic HBV infection (immune tolerance group and immune clearance group) and healthy controls blood separation PMBC, in some cases by immunomagnetic bead technology (IBT) was further purified CD4 ~~ T cells and CD8 ~ T cells incubated with purified using the ELISPOT assay with chronic HBV infection-specific Th1 cell responses and FCM CTL activation ability of IFN-γ secretion in the peripheral blood of infected persons and CD8 ~ T lymphocytes surface CD107 expression Determination judge the ability of CpG ODN activation of the immune cells of chronic HBV infection. Results: HBsAg and M [of HBsAg ~ of CpG ODN] with chronic HBV infection, PBMC, purified CD4 ~ T cells and CD8 ~ T cells were incubated in the comparison of different immune status group: PBMC and purified CD4 ~ T cells to produce IFN-γ number of spots and purified CD8 ~ T lymphocyte surface CD107 expression were no significant differences (P gt; 0.05); immune clearance and normal control group in PBMC CD8 ~ T lymphocytes express CD107 there is a significant difference (P immune clearance group = 0.008 and P healthy control group = 0.007), and CD107a CD107b proportion is much higher than. M stimulation of PBMC purified CD4 ~ T cells produced IFN-γ spots number, and there were significant differences (P = 0.000); the the M stimulus chronic HBV infection PBMC, CD4 ~ T lymphocytes, and CD8 ~ T lymphocytes, between the different immune pairwise comparison: both the PMBC and CD4 ~ T lymphocytes produce IFN-γ spots CD8 ~ T lymphocyte surface CD107 expression were significant differences (P lt; 0.05) ; Furthermore, the purification rate of PBMCs, CD4 ~ T cells and CD8 ~ T cells more than 90%, after purification of the various types of the proportion of dendritic cells are pDC was 30.28%, the MDC-1 to 66.50%, and the MDC-2 was 0.04%. Conclusion: in immune tolerance and the state of immune clearance of chronic HBV infection, CpG ODN can be slightly increased HBsAg stimulation of PBMC, purified CD4 ~~ T lymphocytes secrete IFN-γ and purified CD8 ~~ T lymphocyte surface CD107a and CD107b expression of HBsAg and M was no significant difference in CpG ODN superior stimulating effect on PBMC CD4 ~ T lymphocytes. However, the status of immune clearance, CpGODN can significantly to increase HBsAg stimulate CD8 ~ T lymphocytes in chronic HBV infection in PBMC surface CD107a and CD107b expression. Using immunomagnetic beads separated PBMC in DC has practical significance for the study of the function of purified DC.
