Effect of Parecoxib Preemptive Analgesia on the Recovery Period and Perioperative Cytokine Response in Patients Undergoing Neurosurgery Surgery

Abstract


OBJECTIVE:To investigate the effects of parecoxibe sodium preemptive analgesia on the recovery period and perioperative cytokine response in patients undergoing neurosurgery surgery.METHODS:80 ASAⅠ-Ⅱpatients undergoing neurosurgery surgery were randomly divded into 2 groups. The parecoxib group(group P) received parecoxib 40mg 20 minutes preoperatively and the control group received saline 5ml at the same time. The style of anesthesia were general anesthesia. After the operation all the patients were used patient controlled intravenous analgesia (PCIA). To record the time of extubation and the scores of Ramsay and Agitation in 20minuts after the surgery. To record the VAS score in 2、6、12、24 hour after the surgery and the occurrence of adverse reactions as nausea and vomiting. Venous blood samples were obtained at 10 minutes before Parecoxib or 0.9% saline was injected(T1)、30 minutes(T2) after surgery was initiated and 2h (T3)、4h (T4)、8h (T5)、24h (T6) after the operation for evaluating the level of the cytokine.RESULTS:There were no significant differences in the general conditions between the two groups. The time of extubation was same between the two groups, but the Ramsay scores of Group P was higher than the Group C, the Agitation score of Group P was lower than the Group C (P<0.05).The VAS score in 2h、6h and 12h after the operation of Group P were lower than the Group C. The occurrence of adverse reactions as nausea and vomiting were same between the two groups. The plasma level of TNF-a of Group P was not changed compared with Group C (P>0.05). The plasma levels of IL-6 and IL-8 of Group P were significant lower than the Group C at T3、T4 and T5(P<0.05).CONCLUSION:①Preoperatively-administered parecoxib during neurosurgery surgery can produce better analgesia effect, reduce the VAS scale and the Agitation score.②Preoperatively-administered parecoxib during neurosurgery surgery can reduce the production of cytokines,
