Abstract


Objective:To explore the possible mechanism of isoflurane on learning and memory abilities and hippocampal synapse of offspring rats.Method:Ten healthy Maternal Sprague-Dawle rats in 14 day weighing 220-250g were randomly divided into 2 groups:control group and isoflurane treatment group. The rats of control group were ventilated alone for 2h, the rats of isoflurane treatment group were inhaled isoflurane at the concentration of 1.3% for 2h, The offspring rats of two groups were tested for visual-spatial learning and memory abilities in Morris water maze performed on postnatal 4th week. Then all offspring rats were killed and hippocampus dissociated. The synaptic structure of hippocampus CA1 area was observed, analyzed by trans-electron microscopy.Results:In Morris water maze, the escape latency of control group were significantly prolonged, and the time of spanning flat roof decreased(P<0.05). The number of synapse in hippocampus CA1 area in control group were significantly more than in the isoflurane treatment group, and the structure of hippocampus in control group was integrity, but incomplete in the isoflurane treatment group, meanwhile, the thickness of synaptic density in control group was remarkably increased than that in the isoflurane treatment group.Conclusion:Repeatedly isoflurane exposure may change the synaptic morphology and number, thus affect learning and memory in newborn mice.
