Abstract


ObjectiveTo investigate the Connective Tissue Growth Factor (CTGF) expression before and after pulsed dye laser(PDL,595nm) treatment, and to understand the mechanism of pulsed dye laser treatment of keloid better.Method26 keloid patients were recruited. Two keloids with two similar location, disease duration, texture and appearance were chosen for study, one treated and the other as control.3 sessions of pulsed dye laser treatment were performed on the keloid at 3-week to 4-week interval. Punch biopsy was carried out on both the treated keloid and the control before the first treatment and after the final treatment. The specimen underwent realtime PCR and immunohistochemistry to investigate the mRNA and protein expression of CTGF after PDL treatment.ResultsAccroding to Realtime-PCR, the CTGF mRNA were significantly downregulated after PDL treatment in 96.3% patients as compared to the control group. Immunohistochemistry investigation showed that after treatment the CTGF positive cells also significantly decreased as compared to the control group in 92.59% patients. Vancouver scar scale (VSS) averagely decrease by 20.85±12.33% after treatment.ConclusionsThe pulsed dye laser treatment to keloid can downregulate the expression of CTGF and this might partially explain the mechanism of PDL treatment of keloid.
